| ○ | ○ | Article | A Hybrid RSM-ANN-GA Approach on Optimization of Ultrasound-Assisted Extraction Conditions for Bioactive Component-Rich Stevia rebaudiana (Bertoni) Leaves Extract | Stevia rebaudiana (Bertoni) leaves consist of dietetically important diterpene steviol glycosides (SGs): stevioside (ST) and rebaudioside-A (Reb-A). ST and Reb-A are key sweetening compounds exhibiting a sweetening potential of 100 to 300 times more intense than that of table sucrose. Ultrasound-assisted extraction (UAE) of SGs was optimized by effective process optimization techniques, such as response surface methodology (RSM) and artificial neural network (ANN) modeling coupled with genetic algorithm (GA) as a function of ethanol concentration (X-1: 0-100%), sonication time (X-2: 10-54 min), and leaf-solvent ratio (X-3: 0.148-0.313 g.mL(-1)). The maximum target responses were obtained at optimum UAE conditions of 75% (X-1), 43 min (X-2), and 0.28 g.mL(-1) (X-3). ANN-GA as a potential alternative indicated superiority to RSM. UAE as a green technology proved superior to conventional maceration extraction (CME) with reduced resource consumption. Moreover, UAE resulted in a higher total extract yield (TEY) and SGs including Reb-A and ST yields as compared to those that were obtained by CME with a marked reduction in resource consumption and CO2 emission. The findings of the present study evidenced the significance of UAE as an ecofriendly extraction method for extracting SGs, and UAE scale-up could be employed for effectiveness on an industrial scale. | ○ | ○ | Review | ACCURACY OF REMOVABLE PARTIAL DENTURE METAL FRAMEWORKS FABRICATED BY COMPUTER-AIDED DESIGN/COMPUTER-AIDED MANUFACTURING METHOD: A SYSTEMATIC REVIEW AND META-ANALYSIS | Objective This systematic review and meta-analysis aimed to investigate the accuracy of removable partial denture (RPD) frameworks fabricated by computer-aided de-sign/ computer-aided manufacturing (CAD/CAM) systems compared to frame-works produced by conventional casting methods.Methods A systematic literature search was conducted in electronic databases following the preferred reporting items for systematic reviews and meta-analyses (PRISMA) guidelines, using formulated Boolean operator for searching articles evaluating the fit accuracy of CAD/CAM-fabricated RPD metal frameworks. The agreement of data collection among the reviewers was confirmed using the Cohen kappa coefficient. The modified methodological index for non-randomized studies (MI-NORS) scale was used to assess the quality of the included studies. Heterogene-ity among studies was evaluated, and meta-analyses with global and subgroup analyses were performed.Results A total of 208 articles were identified with 18 articles available for the narrative review and 7 studies available for meta-analysis on the 3D-printed frameworks for RPD (the overall Cohen kappa coefficient = 0.944). There was no article on the accuracy of RPD frameworks made by milling method. The meta-analysis re-vealed that the misfit of frameworks obtained with the CAD/CAM method was significantly higher than that of the conventional lost wax and casting method (SMD = 1.23 gm, 95% CI = 0.3610-2.0981 gm, z = 2.77, P = .0055). Regarding the CAD/CAM techniques used for fabricating RPD metal frameworks, the within subgroup analysis showed that the CAD/CAM-based indirect fabrication method produced frameworks with fit accuracy more similar to the conventional lost-wax and casting method (SMD = 1.15 gm, 95% CI = - 0.06136-2.9192 gm, z = 1.28, P < .01) than the CAD/CAM-based direct fabrication method (SMD = 1.35 gm, 95% CI = -0.2722-2.4381 gm, z = 2.45, P < .01), even though there was no statistical difference between the techniques ( P = .8482).Conclusions The fit accuracy of RPD metal frameworks fabricated by the CAD/CAM method was lower than that of frameworks fabricated by the conventional casting method, but it was within the clinically acceptable range in dimensional misfit. | ○ | ○ | Article | Allulose Attenuated Age-Associated Sarcopenia via Regulating IGF-1 and Myostatin in Aged Mice | Scope Allulose is shown to increase the muscle weight in diet-induced obese mice. However, there are no studies on the effects of allulose in age-associated sarcopenia. This study aims to elucidate the mechanisms of action for allulose in age associated by analyzing the transcriptional patterns in aged mice. Methods and Results The 48-week-old mice are fed with AIN-93diet containing allulose for 12 weeks. Allulose supplementation increases the muscle mass and grip strength in aged mice. Allulose increases the insulin-like growth factor 1 (IGF-1) and its downstream factor expressions which 40 are related protein synthesis, while inhibits the myostatin expression related protein degradation. In mRNA-seq analysis, allulose supplementation significantly decreases in Adiponectin, Adipsin, cell death inducing DFFA like effector (CIDEC), Haptoglobin, Neuroglobin, and stearoyl-CoA desaturase-1 (SCD1) and increases in cytokine-inducible SH2-containing protein (CISH) and ceramide synthase 1 (CerS1) that are regulate protein turn over in gastrocnemius. Also, allulose alleviates autophagy in muscle with regulated mammalian target of rapamycin (mTOR) signaling pathway and increases the anti-oxidant enzyme activity. | ○ | ○ | Article | Crystal structure of multi-functional enzyme FadB from Cupriavidus necator: Non-formation of FadAB complex | Cupriavidus necator H16 is a gram-negative chemolithoautotrophic bacterium that has been extensively studied for biosynthesis and biodegradation of polyhydroxyalkanoate (PHA) plastics. To improve our understanding of fatty acid metabolism for PHA production, we determined the crystal structure of multi-functional enoyl-CoA hydratase from Cupriavidus necator H16 (CnFadB). The predicted model of CnFadB created by AlphaFold was used to solve the phase problem during determination of the crystal structure of the protein. The CnFadB structure consists of two distinctive domains, an N-terminal enol-CoA hydratase (ECH) domain and a C-terminal 3-hydroxyacyl-CoA dehydrogenase (HAD) domain, and the substrate-and cofactor-binding modes of these two functional domains were identified. Unlike other known FadB enzymes that exist as dimers complexed with FadA, CnFadB functions as a monomer without forming a complex with CnFadA. Small angle X-ray scattering (SAXS) measurement further proved that CnFadB exists as a monomer in solution. | ○ | ○ | Article | Cytogenetic assessment of Haworthia using flow cytometry and fluorescence in situ hybridization | The purpose of this study was to investigate cytogenetic variations between the nine Haworthia species necessary for subsequent breeding. Flow cytometric analysis of H. badia 'Murasaki', H. splendens, H. truncata 'Seiko', H. cooperi var. obtusa, H. cymbiformis, H. mirabilis var. mundula, and H. retusa showed a wide range of DNA content from 10.17 to 12.17 pg/1C, whereas two tetraploids, H. limifolia and H. angustifolia var. baylisii, showed higher DNA content of 16.81 and 12.64 pg/1C, respectively. Seven diploid Haworthia expressed three karyotype formulas, whereas two tetraploid Haworthia each expressed a different karyotype formula. Secondary constriction was observed in H. truncata 'Seiko' and H. cooperi var. obtusa. Results of fluorescence in situ hybridization (FISH) showed a major difference between the diploid and tetraploid Haworthia in the distribution of 5S and 18S ribosomal DNA (rDNA) loci. There were eight loci in two out of seven diploids and both tetraploid Haworthia, while H. cymbiformis and H. mirabilis var. mundula each showed seven 18S rDNA loci. H. badia 'Murasaki', H. splendens, and H. retusa had four, three, and five 18S rDNA loci, respectively. In addition, a single pair of 5S rDNA loci was observed in all diploid Haworthia except H. cooperi var. obtusa, which had four 5S rDNA loci. Each tetraploid Haworthia had four 5S rDNA loci, with two strong and two weak signals. All 5S and 18S rDNA loci were observed on the long arm, in the subtelomeric or telomeric region, while a significant variation in chromosome length was observed among the Haworthia. | ○ | ○ | Article | Enzymatic Hydrolysis Modifies Emulsifying Properties of Okra Pectin | Okra pectins (OKPs) with diverse structures obtained by different extraction protocols have been used to study the relationship between their molecular structure and emulsifying properties. A targeted modification of molecular structure offers a more rigorous method for investigating the emulsifying properties of pectins. In this study, three glycoside hydrolases, polygalacturonase (PG), galactanase (GL), and arabinanase (AR), and their combinations, were used to modify the backbone and side-chains of OKP, and the relationships between the pectin structure and emulsion characteristics were examined by multivariate analysis. Enzymatic treatment significantly changed the molecular structure of OKP, as indicated by monosaccharide composition, molecular weight, and structure analysis. GL- and AR- treatments reduced side-chains, while PG-treatment increased side-chain compositions in pectin structure. We compared the performance of hydrolyzed pectins in stabilizing emulsions containing 50% v/v oil-phase and 0.25% w/v pectin. While the emulsions were stabilized by PG (93.3% stability), the emulsion stability was reduced in GL (62.5%), PG+GL+AR (37.0%), and GL+AR (34.0%) after 15-day storage. Furthermore, microscopic observation of the droplets revealed that emulsion destabilization was caused by flocculation and coalescence. Principal component analysis confirmed that neutral sugar side-chains are key for long-term emulsion stabilization and that their structure explains the emulsifying properties of OKP. | ○ | ○ | Article | Evaluation of young F1 Lilium hybrids using fluorescence in situ hybridization analysis and morphological attributes | Lilium, an important ornamental crop, requires genetic and phenotypic assessment in the development of new hybrids. Therefore, genetic (using FISH) and morphological studies were conducted on newly developed hybrids belonging to Lilium genomes (Asiatic, Oriental, longiflorum). Morphological results revealed that all F-1 hybrids showed significant differences from one another. Triploid Asiatic hybrids (Brunello x Mapira) showed good phenotypic characteristics and longiflorum hybrids (Bright Tower x Snow Queen) expressed good growth characters. The Brunello x Mapira hybrid was especially superior in root length, number of scales, bulb diameter and bulb weight. Chromosomal studies confirmed that all hybrids were diploid (2x = 24) with 24 chromosomes except the "AA-2" Asiatic hybrid which was triploid (2x = 3x = 36). In our karyotyping results, there was non-significant variation with respect to short arm length and arm length ratio among hybrids, while long arm length and total chromosome length showed significant differences. In the case of 45S signals, the maximum number of 45S rDNAs were found in "Bright Tower x Snow Queen" while the minimum number of 45S signals (2) were found in "Yellow cocotte x Valdisole". 5S signals were similar in all hybrids except triploid Asiatic hybrids. Thus, newly developed young hybrids can be successfully screened using morphological and FISH analysis. | ○ | ○ | Article | Fermented Oyster (Crassostrea gigas) Extract Cures and Prevents Prednisolone-Induced Bone Resorption by Activating Osteoblast Differentiation | Osteoporosis is a bone resorptive disease characterized by the loss of bone density, causing an increase in bone fragility. In our previous study, we demonstrated that gamma aminobutyric acid-enriched fermented oyster (Crassostrea gigas) extract (FO) stimulated osteogenesis in MC3T3-E1 preosteoblast cells and vertebral formation in zebrafish. However, the efficacy of FO in prednisolone (PDS)-induced bone resorption remains unclear. In this study, we evaluated the osteogenic potential of FO in MC3T3-E1 preosteoblast cells and zebrafish larvae under both PDS-pretreated and PDS-post-treated conditions. We found that FO recovered osteogenic activity by upregulating osteoblast markers, such as alkaline phosphatase (ALP), runt-related transcription factor 2, and osterix, in both PDS-pretreated and post-treated MC3T3-E1 osteoblast cells and zebrafish larvae. In both conditions, PDS-induced decrease in calcification and ALP activity was recovered in the presence of FO. Furthermore, vertebral resorption in zebrafish larvae induced by pretreatment and post-treatment with PDS was restored by treatment with FO, along with the recovery of osteogenic markers and downregulation of osteoclastogenic markers. Finally, whether FO disturbs the endocrine system was confirmed according to the Organization for Economic Cooperation and Development guideline 455. We found that FO did not stimulate estrogen response element-luciferase activity or proliferation in MCF7 cells. Additionally, in ovariectomized mice, no change in uterine weight was observed during FO feeding. | ○ | ○ | Article | Lariciresinol Displays Anti-Diabetic Activity through Inhibition of α-Glucosidase and Activation and Enhancement of Insulin Signaling | Scope: The aim of this study is to investigate the antidiabetic effect of lariciresinol (LSR) in C2C12 myotubes and streptozotocin (STZ)-induced diabetic mice. Methods and results: To investigate antidiabetic potential of LSR, alpha-glucosidase inhibitory assay, molecular docking, glucose uptake assay, western blot assay on antidiabetic biomarkers are performed. STZ-induced diabetic model is used for in vivo study by calculating oral glucose tolerance test, histochemical examination, and glycogen assay. LSR inhibits alpha-glucosidase activity with an IC50 value of 6.97 +/- 0.37 mu M and acts as a competitive inhibitor with an inhibitory constant (Ki) value of 0.046 mu M. In C2C12 cells, LSR activates insulin signaling leading to glucose transporter 4 (GLUT4) translocation and augmented glucose uptake. Furthermore, in Streptozotocin (STZ)-treated diabetic mice, 3 weeks of oral LSR administration (10 mg kg(-1)) considerably decrease blood glucose levels, while increasing insulin levels in an oral glucose tolerance test, improve pancreatic islet size, increase GLUT4 expression, and significantly enhance insulin signaling in skeletal muscle. LSR treatment also activates glycogen synthase kinase 3 beta (GSK-3 beta) resulting in improved glycogen content. | ○ | ○ | Article | Optimization of a Multi-Residue Analytical Method during Determination of Pesticides in Meat Products by GC-MS/MS | In this study, a multi-residue analysis was developed for 32 compounds, including pesticides and metabolites, in five meat products using gas chromatography-tandem mass spectrometry (GC-MS/MS). The validation of the developed analytical method was also evaluated in accordance with Codex Alimentarius guidelines. Aminopropyl (NH2), C-18, and florisil solid phase extraction (SPE) cartridges were used to evaluate and optimize the cleanup procedure of the tested samples prior to GC-MS/MS analysis. Based on the analytical performance, the C-18 SPE cartridge was deemed to be the most suitable among the examined SPE cartridges. The optimized method demonstrated that 29 out of 32 tested compounds acquired good linearity (R-2 >= 0.99), and 25 tested compounds displayed the method limit of quantification (MLOQ) <= 0.01 mg/kg. Out of the 32 tested compounds, only 21 compounds met the acceptable analytical criteria for the lard and tallow samples, compared to 27 compounds in the beef, pork, and chicken samples that falls within the acceptable standards for recovery (70-120%) and analytical precision (relative standard deviation RSD <= 20%). The average matrix effect was widely varied (20.1-64.8%) in the studied meat samples that were affected by either ion enhancement or suppression. In particular, in the lard sample, 13 compounds showed poor recovery and analytical precision due to ion suppression. Thus, the matrix effect (ME) was considered a critical factor during multi-residue pesticide analysis in different meat products. In conclusion, this developed analytical method can be used as a routine monitoring system for residual pesticide analysis in livestock products with acceptable analytical standards. | ○ | ○ | Article | Plum Prevents Intestinal and Hepatic Inflammation in the Acute and Chronic Models of Dextran Sulfate Sodium-Induced Mouse Colitis | Scope Inflammatory bowel disease (IBD), including ulcerative colitis (UC), is a chronic recurrent inflammatory disease of the digestive tract and increases the risk of colon cancer. Method and Results This study evaluates the effects of dietary intervention with freeze-dried plum (FDP), a natural antioxidant and anti-inflammatory fruit with no toxicity on dextran sulfate sodium (DSS)-induced acute and chronic experimental colitis in a mouse model and studies the molecular mechanisms of protection through the gut-liver axis. The results show that FDP decreases the levels of inflammatory mediators, which is a nitrative stress biomarker in both acute and chronic models. FDP markedly reduces DSS-induced injury to the colonic epithelium in both acute and chronic models. In addition, FDP significantly decreases the levels of pro-oxidant markers such as CYP2E1, iNOS, and nitrated proteins (detected by anti-3-NT antibody) in DSS-induced acute and chronic colonic injury models. Furthermore, FDP markedly reduces markers of liver injury such as serum ALT/AST, antioxidant markers, and inflammatory mediators in DSS-induced acute and chronic colonic injury. | ○ | ○ | Article | Prediction of strawberry fruit yield based on cultivar-specific growth models in the tunnel-type greenhouse | The strawberry growth and fruit yield of five Korean cultivars in the tunnel-type greenhouse predicted using their growth. The number of leaves, petiole length, leaf length and width, crown diameter, and the ratio of red and far-red (RFR) of the five Korean cultivars were measured during the cultivation period. The number of leaves of all cultivars exhibited a similar trend during this period; the plant and petiole length of 'Maehyang' were the longest, leaf length exhibited similar trends in all five cultivars except for 'Jukhyang', the leaf width of 'Arihyang', was the longest, and crown diameter of 'Keumsil' was the thickest. The leaf length, crown diameter, and RFR were associated with the fruit yield in the multiple linear regression. When a single model was used to predict the yield of all five cultivars, the correlation between expected yield and actual yield was r = 0.53. When cultivar-specific models were built for the prediction, the correlation increased to r = 0.77. | ○ | ○ | Article | Multiparticle ⁿatSr(γ, xnyp) reactions induced with bremsstrahlung end-point energies of 55, 60, and 65 MeV | This study measured the yields of the radionuclides Sr-82, Sr83(m+g), Sr-85m, Sr-85g, Sr-87m, Rb81(g+0.976m),Rb-82m,Rb-83g,Rb84(m+g), and Rb86(m+g) produced in Sr-nat(gamma, xnyp) multiparticle reactions with bremsstrahlung end-point energies of 55, 60, and 65 MeV. The bremsstrahlung radiation was generated using the 100-MeV electron linear accelerator at the Pohang Accelerator Laboratory, Korea, and the reaction yields were derived from the induced activities-measured using off-line gamma-ray spectrometry. To obtain accurate experimental results, we performed the necessary gamma-ray interference corrections. The experimental results were compared with the theoretical predictions obtained usingthe TALYS-1.95 statistical nuclear model code. The calculations were performed using six different level-density models to ascertain which model best fitted the experimental results. | ○ | ○ | Article | Semi-Transparent, Micrometer Resolution p-NiO/n-ZnO Heterojunction Diode Temperature Sensors with Ultrathin Metal Anode | Various temperature sensitive biological mechanisms have been utilized for new biomedical engineering tools such as neuromodulation, cancer cell hyperthermia or photothermal therapy. Optically transparent and high spatio-temporal resolution temperature sensors are needed to precisely analyze the biological effects that occur in response to the temperature changes. In this work, semi-transparent p-NiO/n-ZnO heterojunction diode-based temperature sensors with 100 mu m-diameter ultrathin transparent Au/Ag metal anodes is introduced. The fabricated diode temperature sensors accurately measure temperature changes from 25 to 80 degrees C, which is of significant interest in many biomedical engineering applications. The sensors also exhibit adequate transparency over the entire visible light spectrum for biomedical imaging including fluorescent microscopy. Low-power operation of the temperature sensor (<0.2 nW) is achieved to avoid a self-heating effect. | ○ | ○ | Article | Adaptive optics and VLBA imaging observations of recoiling supermassive black hole candidates | We present the results of high-resolution adaptive optics imaging observations of four kinematically identified recoiling supermassive black hole (rSMBH) candidates. Ellipse fitting was carried out to measure the spatial offset between the active galactic nucleus (AGN) and the centre of the host galaxy. Two rSMBH candidates (J1713 + 3523 and J2054 + 0049) are found to be offset AGN. However, the Very Long Baseline Array 1.5 GHz continuum imaging observation and spectral decomposition of the [O iii]5007 line suggest that J1713 + 3523 is a dual AGN and its spatial offset is not due to a recoil event. The spatial offset between the AGN and the centre of the host galaxy in J2054 + 0049 is 0.06 +/- 0.01 arcsec (201 +/- 22 pc).
