| ○ | ○ | Article | Design, fabrication, and characterization of piezoelectric single crystal stack actuators based on PMN-PT | A piezoelectric stack actuator (PSA) consisting of 28 lead magnesium niobate-lead titanate (PMN-PT) piezoelectric single crystal layers is developed. The free stroke and blocking force, which are the figures of merit of a PSA, are simulated through finite element analysis. The free stroke of the PSA samples measured using a laser displacement sensor was in the range of 6.4-8.0 mu m at 200 V. In addition, the blocking force of the PSA samples measured using a custom-made system, consisting of a strain gauge, a motor and a load cell, was in the range of 191-226 N at 200 V. | ○ | ○ | Article | Determination of homogenized material constants of the 2-2 mode piezocomposite | 2-2 mode piezocomposites are widely used for the drive section of acoustic transducers. Typically, piezocomposites have been developed using theoretical or numerical methods such as the finite element method. However, rigorous analysis of piezocomposites has always been difficult due to their complicated two-phase structure, which necessitates homogenization of the structure with a single-phase material with properties identical to those of the original two-phase piezocomposite. In this study, we determined the material properties of the equivalent single-phase material using the resonator method. The asymptotic averaging method was used to obtain the initial properties of the homogenized material, which had some accuracy limitations. We improved the accuracy by using multiple resonators, each of which reflected the effect of different material constants. We adjusted the material constants that influence each resonator to minimize the difference between the original piezocomposite's resonant and antiresonant frequencies and those of the equivalent single-phase material. The optimization technique was used to facilitate this adjustment. The homogenized material with the optimized material constants represented the original two-phase piezocomposite very well. | ○ | ○ | Article | Histopathological Features of Myxoid Pleomorphic Liposarcoma in an African Pygmy Hedgehog (Atelerix Albiventris) | Liposarcoma is a malignant tumor that is derived from fat tissue. Liposarcoma is one of the most common malignant tumors in humans and animals. Liposarcomas are classified into six groups according to the WHO: dedifferentiated liposarcoma, myxoid liposarcoma, pleomorphic liposarcoma, liposarcoma not otherwise specified, atypical spindle cell/pleomorphic lipomatous and myxoid pleomorphic liposarcoma. Each subtype shows distinct pathological features, responses to chemotherapy and/or radiotherapy, and clinical outcomes. In this case, a myxoid pleomorphic liposarcoma was observed in a 5-year-old male African pygmy hedgehog. Myxoid pleomorphic liposarcoma is characterized by pathological features of both pleomorphic liposarcoma and myxoid liposarcoma. The tumor consisted of similar to 60% of the myxoid substance area and similar to 30% of pleomorphic neoplastic cells. The tumor presented similar clinical outcomes, including metastasis and prognosis, to myxoid liposarcoma rather than pleomorphic liposarcoma. This suggests that analyzing the proportion of pleomorphic area and myxoid area in myxoid liposarcoma could provide important information for predicting clinical outcomes. Myxoid pleomorphic liposarcoma is characterized by pathological features of both pleomorphic liposarcoma and myxoid liposarcoma, as the name suggests. In this case, a myxoid pleomorphic liposarcoma was observed in a 5-year-old male African pygmy hedgehog. It consisted of similar to 60% of the myxoid substance area with proliferating round cells and similar to 30% of pleomorphic neoplastic cells. The subject presented with extrapulmonary metastasis, but a good prognosis during 6 months of follow-up, which is similar to the characteristics of myxoid liposarcoma. The histopathological features of myxoid pleomorphic liposarcoma may reflect the features of either myxoid liposarcoma or pleomorphic liposarcoma depending on the proportion of each histopathological feature. The proportion of the pleomorphic area and the myxoid area may offer information on the prognosis and metastasis of myxoid pleomorphic liposarcoma, which will be helpful for setting up a treatment plan. Thus, analyzing the proportion of pleomorphic area and myxoid area could be suggested as one of the ways to predict clinical outcomes. | ○ | ○ | Article | Histopathological Findings and Metagenomic Analysis of Esophageal Papillary Proliferation Identified in Laying Broiler Breeders | Simple Summary This study describes the histopathological finding of mucosal papillary proliferation observed in the esophagus of chicken and attempts to determine its infectious etiology using metagenomics. Unlike squamous metaplasia of the esophageal mucosa due to vitamin A deficiency, the stratified squamous epithelial cells of the esophagus were completely replaced by increased numbers of ducts/ductules, lymphocytes, and connective tissue, and multiple hyperplasia of the esophageal gland was observed. As the result of the metagenomic analysis of esophagus samples from this chicken, no viral cause was identified; however, the contributing role of Bradyrhizobium sp. could not be excluded. In this study, we report the first histopathological examination of a rare case of an esophageal papillary lesion appearing as adenoma in a chicken and highlight the importance of histopathological results for a definitive diagnosis of such cases. White or pale-yellow nodules 2-7 mm in length were observed in the esophageal lumen in a number of laying broiler breeders with reduced laying rates. Metaplasia of the mucosal epithelial layer and mucous gland, as well as lymphocyte infiltration under the esophageal mucous gland and epithelial cell layer, were observed, which we found were caused by vitamin A deficiency. In one chicken, however, the stratified squamous epithelial cells of the esophagus were completely replaced by increased numbers of ducts/ductules, lymphocytes, and connective tissue, resulting in a papillary morphology. The ducts were surrounded by a fibrous stroma. Multiple hyperplasia of the esophageal gland was also observed and the esophageal glands were lined by a single layer of columnar cells, and a large number of lymphocytes were infiltrated into the submucosal layer. Based on the gross findings, this papillary proliferation was considered papilloma, but histopathologically, a mass composed of squamous epithelium was not observed. Therefore, the papillary lesion appeared as adenoma with squamous metaplasia of the esophageal gland and ectasia, or mucosal epithelial papillary hyperplasia, associated with chronic esophagitis. A metagenomic analysis of the esophagus samples from this chicken was performed to determine the infectious etiology. | ○ | ○ | Article | Postoperative Observation of Spaying with the Silicon Ring on the Ovaries in Heifers: A Retrospective Study in 28 Cases | Simple Summary Because the estrus in cattle is thought to be energy-consuming if the animals are not planned to breed, spaying in cattle has recently been applied to improve daily weight gain and meat quality. However, postoperative changes in ligation-spaying method with the silicon ring to ovaries via transvaginal methods in heifers have not been clearly identified. This retrospective study presented that heifers spayed with this method exhibited no estrus signs at the pubertal age and the ligated ovaries disappeared within a month post-surgery due to ischemic necrosis. Whereas ovarian steroid hormone levels in spayed heifers were not changed following the puberty, luteinizing hormone level at the pubertal age was higher than unspayed controls. Although carcass weight and yield were similar between groups at the pubertal age upon slaughtering, the spayed animals presented higher marbling degree than that of controls. These results may contribute to develop new management strategies for livestock. Although spaying prepubertal heifers has routinely been conducted to control cattle herd and improve meat quality, understandings of the postoperative changes following new spaying methods with the silicon ring on the ovaries via colpotomy remain limited. Therefore, as a retrospective study, 28 cases of spayed heifers were reviewed for postoperative changes after employing this method, with inclusion criteria including complete medical records for clinical observation, ultrasonography, measuring reproductive hormones, and tracking slaughter records. No mortality and heat signs at the pubertal age postoperatively occurred in spayed animals. On ultrasonography during rectal examination, the ovaries were enlarged without any folliculogenesis from one week, while massive ovarian edema appeared from two weeks, and ovaries were no longer palpable at four weeks post-surgery. In hormones, whereas estrogen and progesterone levels did not change from prepubertal to pubertal age in spayed animals, luteinizing hormone levels progressively increased during this period and reached a higher level at pubertal period than unspayed controls. Although carcass weight and yield were similar between groups upon slaughter at pubertal age, the spayed animals presented higher carcass quality (marbling degree) than that of controls. | ○ | ○ | Article | Thermal performance enhancement of shell and helical coil heat exchanger using MWCNTs/water nanofluid | Heat exchangers serve as pivotal components in industries. Enhancement in the performance of heat exchangers has been the focus of researchers in the recent past. The aim of this study was to numerically investigate the improvement in thermal performance of heat exchangers by combining two passive techniques, i.e., use of nanofluids and coiled tubes. Thermohydraulic performance of a shell and helical coil tube heat exchanger was evaluated at 0.1%, 0.3% and 0.5% volume concentrations of MWCNT/water nanofluid and at coil Reynolds numbers of 9000-15,000. The numerical simulations were performed considering the cold-fluid (water) flows through the shell, whereas the hot fluid (water and nanofluid) flows through the helical coil in a counter-current flow direction. The RNG k-epsilon model with enhanced wall treatment was applied. The results illustrate that the Nusselt number of the fluid flowing through the coil increased as coil Reynolds number and nanofluid volume concentration increased. When multiwall carbon nanotube/water nanofluid with a volume concentration of 0.5% was used at a coil Reynolds number of 15,000, the highest improvement in Nusselt number was observed i.e., 31.5% and the highest pressure drop of 47.35% was noted at the same conditions The maximum decline of 40.5% in the effectiveness of the heat exchanger was found when Reynolds number increased from 9000 to 15,000 at 0.5% volume concentration of nanofluid. | ○ | ○ | Article | Accelerating NADH oxidation and hydrogen production with mid-gap states of nitrogen-rich carbon nitride photocatalyst | Regeneration of electron carriers such as NAD(+)/NADH is highly desirable and essential for enzymatic conversions. Here, we demonstrate a sustainable strategy for the regeneration of NAD(+) as an electron carrier via photon-assisted heterogeneous catalysis. For this, a mid-gap state induced nitrogen-rich polymeric carbon nitride (NPCN) catalyst was synthesized by an additive-assisted thermal copolymerization. Utilizing NPCN as a photocatalyst presented NADH photooxidation efficiency of over 98% and a high hydrogen production rate of 11.18 mmolg(-1)h(-1) with an apparent quantum yield of 9.16% (lambda = 420 nm), outperforming other state-of-art metal-free photocatalysts. | ○ | Article | Cas9-mediated gene editing in the black-legged tick, Ixodes scapularis, by embryo injection and ReMOT Control | Despite their capacity to acquire and pass on an array of debilitating pathogens, research on ticks has lagged behind other arthropod vectors, such as mosquitoes, largely because of challenges in applying available genetic and molecular tools. CRISPR-Cas9 is transforming non-model organism research; however, successful gene editing has not yet been reported in ticks. Technical challenges for injecting tick embryos to attempt gene editing have further slowed research progress. Currently, no embryo injection protocol exists for any chelicerate species, including ticks. Herein, we report a successful embryo injection protocol for the black-legged tick, Ixodes scapularis, and the use of this protocol for genome editing with CRISPR-Cas9. We also demonstrate that the ReMOT Control technique could be successfully used to generate genome mutations outside Insecta. | ○ | ○ | Article | Clusterin is involved in mediating the metabolic function of adipose SIRT1 | SIRT1 is a metabolic sensor regulating energy homeostasis. The present study revealed that mice with selective overexpression of human SIRT1 in adipose tissue(Adipo-SIRT1) were protected from high-fat diet (HFD)-induced metabolic abnormalities.Adipose SIRT1 was enriched at mitochondria-ER contacts (MERCs) to trigger mitohormesis and unfolded protein response (UPRmt), in turn preventing ER stress. As a downstream target of UPRmt, clusterin was significantly upregulatedand acted together with SIRT1 to regulate the protein and lipid compositions at MERCs of adipose tissue. In mice lacking clusterin, HFD-induced metabolic abnormalities were significantly enhanced and could not be prevented by overexpression of SIRT1 in adipose tissue. Treatment with ER stress inhibitors restored adipose SIRT1-mediated beneficial effects on systemic energy metabolism. | ○ | ○ | Article | Genome structure and evolutionary history of frankincense producing Boswellia sacra | Boswellia sacra Flueck (family Burseraceae) tree is wounded to produce frankincense. We report its de novo assembled genome (667.8 Mb) comprising 18,564 high-confidence protein-encoding genes. Comparing conserved single-copy genes across eudicots suggest >97% gene space assembly of B. sacra genome. Evolutionary history shows B. sacra gene-duplications derived from recent paralogous events and retained from ancient hexaploidy shared with other eudicots. The genome indicated a major expansion of Gypsy retroelements in last 2 million years. The B. sacra genetic diversity showed four clades intermixed with a primary genotype-dominating most resin-productive trees. Further, the stem transcriptome revealed that wounding concurrently activates phytohormones signaling, cell wall fortification, and resin terpenoid biosynthesis pathways leading to the synthesis of boswellic acid-a key chemotaxonomic marker of Boswellia. | ○ | ○ | Article | Oxygen migration energy in La and Y Co-doped CeO2: Effect of lattice constant and grain boundary segregation | The transfer energy of oxygen vacancy of La and Y co-doped CeO2 is investigated in which the total amount of La and Y is fixed at 20 at%. Although both the La3+ (1.16 angstrom) and Y3+ (1.032 angstrom) ions are larger than the CO4+ ions (0.97 angstrom), individual single doping of La and Y results in an increase and decrease in the CeO2 lattice constant, respectively, due to the effect of the critical ionic radius (r(c)). As the La/Y ratio varies, the lattice constant increases as the La content increases, which leads to a decrease in the migration energy of the oxygen vacancy in the grain interior. When La and Y are co-doped at a ratio of 2:18, its lattice constant is almost the same as that of pure CeO2 in which the dissociation energy of the oxygen vacancy achieves the lowest value due to the minimum lattice-strain energy. The migration energy at the grain boundary also decreases with increasing La content. This result is explained by the degree of grain boundary segregation of the dopant. | ○ | ○ | Article | Effect of Composite Chitosan/Sodium Alginate Gel Coatings on the Quality of Fresh-Cut Purple-Flesh Sweet Potato | In this study, single-layer coating using chitosan (Ch) and sodium alginate (SA) solutions and their gel coating (ChCSA) formed by layer-by-layer (LbL) electrostatic deposition using calcium chloride (C) as a cross linking agent were prepared to improve storage qualities and shelf-life of fresh-cut purple-flesh sweet potatoes (PFSP). The preservative effects of single-layer coating in comparison with LbL on the quality parameters of fresh-cut PFSP, including color change, weight loss, firmness, microbial analysis, CO2 production, pH, solid content, total anthocyanin content (TAC), and total phenolic content (TPC) were evaluated during 16 days of storage at 5 degrees C. Uncoated samples were applicable as a control. The result established the effectiveness of coating in reducing microbial proliferation (similar to 2 times), color changes (similar to 3 times), and weight loss (similar to 4 times) with negligible firmness losses after the storage period. In addition, TAC and TPC were better retained in the coated samples than in the uncoated samples. In contrast, quality deterioration was observed in the uncoated fresh cuts, which progressed with storage time. | ○ | ○ | Article | Effect of Low-Dose Persistent Organic Pollutants on Mitochondrial Function: Human and in Vitro Evidence | Background: Chronic exposure to low-dose persistent organic pollutants (POPs) can induce mitochondrial dysfunction. This study evaluated the association between serum POP concentrations and oxygen consumption rate (OCR) as a marker of mito-chondrial function in humans and in vitro cells. Methods: Serum concentrations of organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) were measured in 323 adults. The OCRs of platelets and peripheral blood mononuclear cells (PBMCs) were assessed in 20 mL of fresh blood using a Seahorse XF analyzer. Additionally, the in vitro effects of Arochlor-1254, beta-hexachlorocyclohexane, and p,p '-dichlorodiphenyltrichloroethane at concentrations of 0.1 pM to 100 nM were evaluated in human platelets, human PBMCs, and Jurkat T-cells. Results: The association between serum POP concentrations and OCR differed depending on the cell type. As serum OCP con-centrations increased, basal platelet OCR levels decreased significantly; according to the OCP quintiles of summary measure, they were 8.6, 9.6, 8.2, 8.0, and 7.1 pmol/min/mu g (P trend= 0.005). Notably, the basal PBMC OCR levels decreased remarkably as the serum PCB concentration increased. PBMC OCR levels were 46.5, 34.3, 29.1, 16.5, and 13.1 pmol/min/mu g according to the PCB quintiles of summary measure (P trend <0.001), and this inverse association was consistently observed in all subgroups stratified by age, sex, obesity, type 2 diabetes mellitus, and hypertension, respectively. In vitro experimental studies have also dem-onstrated that chronic exposure to low-dose POPs could decrease OCR levels.
