| ○ | ○ | Article | Exosomal Del-1 as a Potent Diagnostic Marker for Breast Cancer: Prospective Cohort Study | Currently, there are no molecular biomarkers for the early detection of breast cancer. Our previous study identified surface proteins found on circulating exosomes for detecting early-stage breast cancer. In the current study, we aimed to confirm the diagnostic role of exosomal developmental endothelial locus-1 (Del-1) in a prospective cohort with breast cancer by comparing plasma exosomal Del-1 levels before and after curative surgery. This study confirmed the normalization of exosomal Del-1 after surgery, indicating exosomal Del-1 as a potent diagnostic biomarker for breast cancer. In addition, because a high Del-1 level after surgery was associated with early relapse, this suggests exosomal Del-1 as a potential prognostic marker by identifying the existence of residual cancer. Background: The differential diagnostic role of plasma developmental endothelial locus-1 (Del-1) was proposed in our previous study. Therefore the current study aimed to confirm the diagnostic role and explore the prognostic role of exosomal Del-1 in a prospective cohort of female patients with breast cancer. Patients and Methods: To determine the optimal sampling time for the postoperative Del-1 measurements, blood was serially collected on days 1, 3, 5, and 7 after surgery in 22 patients (cohort 1). Thereafter, 111 female patients with breast cancer were prospectively enrolled (cohort 2) to compare exosomal Del-1 levels before and after surgery. Results: Among the subsequent prospective cohort, 107 patients (96.4%) showed a high exosomal Del-1 level (optical density [OD] value > 0.5) at the time of diagnosis. Of these patients, 101 (94.6%) in this high-level group showed normalized Del-1 levels postoperatively, representing a significant difference (mean OD value, 1.232 vs. 0.196; P < .00001). High postoperative Del-1 level was significantly associated with a worse disease-free survival adjusted to the clinicopathological characteristics (hazard ratio, 24.0; P = .0011). Conclusion: This study confirmed the normalization of exosomal Del-1 after surgery, indicating exosomal Del-1 as a potent diagnostic biomarker for breast cancer. In addition, because a high Del-1 level after surgery was associated with early relapse, this suggests exosomal Del-1 as a potential prognostic marker by identifying the existence of residual cancer. | ○ | ○ | Article | Meta-analysis of transanal versus laparoscopic total mesorectal excision for rectal cancer: a 'New Health Technology'assessment in South Korea | Purpose: Under the South Korea's unique health insurance structure, any new surgical technology must be evaluated first by the government in order to consider whether that particular technology can be applied to patients for further clinical trials as categorized as 'New Health Technology,' then potentially covered by the insurance sometime later. The aim of this meta-analysis was to assess the safety and efficacy of transanal total mesorectal excision (TaTME) for rectal cancer, activated by the National Evidence-based Healthcare Collaborating Agency (NECA) TaTME committee. Methods: We systematically searched Ovid-MEDLINE, Ovid-Embase, Cochrane, and Korean databases (from their inception until August 31, 2019) for studies published that compare TaTME with laparoscopic total mesorectal excision (LaTME). End-points included perioperative and pathological outcomes. Results: Sixteen cohort studies (7 for case-matched studies) were identified, comprising 1,923 patients (938 TaTMEs and 985 LaTMEs). Regarding perioperative outcomes, the conversion rate was significantly lower in TaTME (risk ratio, 0.19; 95% confidence interval, 0.11-0.34; P < 0.001); whereas other perioperative outcomes were similar to LaTME. There were no statistically significant differences in pathological results between the 2 procedures. Conclusion: Our meta-analysis showed comparable results in preoperative and pathologic outcomes between TaTME and LaTME, and indicated the benefit of TaTME with low conversion. Extensive evaluations of well-designed, multicenter randomized controlled trials are required to come to unequivocal conclusions, but the results showed that TaTME is a potentially beneficial technique in some specific cases. This meta-analysis suggests that TaTME can be performed for rectal cancer patients as a 'New Health Technology' endorsed by NECA in South Korea. | ○ | ○ | Article | Differences in seroprevalence between epicenter and non-epicenter areas of the COVID-19 outbreak in South Korea | To compare the standardized severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) seroprevalence of high epicenter region with non-epicenter region, serological studies were performed with a total of 3,268 sera from Daegu City and 3,981 sera from Chungbuk Province. Indirect immunofluorescence assay (IFA) for SARS-CoV-2 IgG results showed a high seroprevalence rate in the Daegu City (epicenter) compared with a non-epicenter area (Chungbuk Province) (1.27% vs. 0.91%, P = 0.0358). It is noteworthy that the highest seroprevalence in Daegu City was found in elderly patients (70's) whereas young adult patients (20's) in Chungbuk Province showed the highest seroprevalence. Neutralizing antibody (NAb) titers were found in three samples from Daegu City (3/3, 268, 0.09%) while none of the samples from Chungbuk Province were NAb positive. | ○ | ○ | Article | Screening of small molecules attenuating biofilm formation of Acinetobacter baumannii by inhibition of ompA promoter activity | Anti-virulence therapeutic strategies are promising alternatives against drug-resistant pathogens. Outer membrane protein A (OmpA) plays a versatile role in the pathogenesis and antimicrobial resistance of Acinetobacter baumannii. Therefore, OmpA is an innovative target for anti-virulence therapy against A. baumannii. This study aimed to develop a high-throughput screening (HTS) system to discover small molecules inhibiting the ompA promoter activity of A. baumannii and screen chemical compounds using the bacterial growth-based HTS system. The ompA promoter and open reading frame of nptI fusion plasmids that controlled the expression of nptI encoding resistance to kanamycin by the ompA promoter were constructed and then transformed into A. baumannii ATCC 17978. This reporter strain was applied to screen small molecules inhibiting the ompA promoter activity in a chemical library. Of the 7,520 chemical compounds, 15 exhibited >= 70% growth inhibition of the report strain cultured in media containing kanamycin. Three compounds inhibited the expression of ompA and OmpA in the outer membrane of A. baumannii ATCC 17978, which subsequently reduced biofilm formation. In conclusion, our reporter strain is useful for large-scale screening of small molecules inhibiting the ompA expression in A. baumannii. | ○ | ○ | Article | The putative sensor histidine kinase VadJ coordinates development and sterigmatocystin production in Aspergillus nidulans | The VosA-VelB heterocomplex governs expression of several genes associated with fungal development and secondary metabolism. In this study, we have investigated the functions of one of the VosA-VelB-activated developmental genes vadJ in development and production of the mycotoxin sterigmatocystin in the model fungus Aspergillus nidulans. The vadJ gene is predicted to encode a 957-amino acid length protein containing a highly conserved sensor histidine kinase domain. The deletion of vosA or velB resulted in decreased mRNA levels of vadJ throughout the life cycle, suggesting that VosA and VelB are necessary for proper expression of vadJ. Nullifying vadJ led to highly restricted colony growth, lowered formation of asexual spores, and about two-fold reduction in conidial viability. Conversely, the deletion of vadJ resulted in elevated production of sexual fruiting bodies and sterigmatocystin. These suggest that VadJ is necessary for proper coordination of asexual and sexual development, and sterigmatocystin production. In accordance with this idea, the deletion of vadJ led to elevated mRNA levels of the two key sexual developmental activators esdC and nsdD. | ○ | ○ | Review | Are Self-Driving Vehicles Ready to Launch? An Insight into Steering Control in Autonomous Self-Driving Vehicles | In the last couple of years, academia-industry collaboration has demonstrated rapid advancements in the development of self-driving vehicles. Since it is evident that self-driving vehicles are going to reshape the traditional transportation systems in near future through enhancement in safe and smart mobility, motion control in self-driving vehicles while performing driving tasks in a dynamic road environment is still a challenging task. In this regard, we present a comprehensive study considering the evolution of steering control methods for self-driving vehicles. Initially, we discussed an insight into the traditional steering systems of the vehicles. To the best of our knowledge, currently, there is no taxonomy available, which elaborates steering control methods for self-driving vehicles. In this paper, we present a novel taxonomy including different steering control methods which are categorized into deterministic and heuristic steering control methods. Concurrently, the abovementioned techniques are critically reviewed elaborating their strengths and limitations. | ○ | Article | Changes in serum protein electrophoresis profiles and acute phase proteins in calves with diarrhea | Calf diarrhea leads to substantial economic losses in the livestock industry worldwide due to medical treatment costs, retarded growth performance, and even death. The objective of this study was to investigate changes in serum protein profiles and acute phase proteins in calves with diarrhea and identify the association between these changes and diarrhea. A total of 185 Korean beef calves were used and divided into 3 groups by age: 1 to 10 days (n = 46), 11 to 20 days (n = 65), and 21 to 30 days (n = 74). Blood and fecal samples were collected from each calf. Serum concentrations of total protein, protein fractions (albumin, alpha 1-globulin, alpha 2-globulin, beta-globulin, and gamma-globulin), haptoglobin (Hp), and serum amyloid A (SAA) were analyzed. Compared to calves without diarrhea, calves with diarrhea had significantly lower albumin concentrations at 11 to 20 days and 21 to 30 days of age (P = 0.017 and P = 0.000, respectively) and significantly higher alpha 1-globulin fractions at 21 to 30 days of age (P = 0.01). Interestingly, alpha 2-globulin fractions were significantly higher in diarrheic calves in all age groups, whereas gamma-globulin fractions were significantly lower in calves with diarrhea aged 1 to 10 days, compared with normal animals. In calves with diarrhea, the concentration of Hp was significantly higher, whereas SAA levels were not different between normal and diarrheic calves. In addition, a positive correlation was found between alpha 2-globulin and Hp (P = 0.0004). | ○ | ○ | Article | Effect of aerodynamic modifications on the surface pressure patterns of buildings using proper orthogonal decomposition | This study analyzed the pressure patterns and local pressure of tall buildings with corner modifications (recessed and chamfered corner) using wind tunnel tests and proper orthogonal decomposition (POD). POD can distinguish pressure patterns by POD mode and more dominant pressure patterns can be found according to the order of POD modes. Results show that both recessed and chamfered corners effectively reduced wind-induced responses. Additionally, unique effects were observed depending on the ratio of corner modification. Tall building models with recessed corners showed fluctuations in the approaching wind flow in the first POD mode and vortex shedding effects in the second POD mode. With large corner modification, energy distribution became small in the first POD mode, which shows that the effect of the first POD mode reduced. Among building models with chamfered corners, vortex shedding effects appeared in the first POD mode, except for the model with the highest ratio of corner modifications. The POD confirmed that both recessed and chamfered corners play a role in reducing vortex shedding effects, and the normalized power spectral density peak value of modes showing vortex shedding was smaller than that of the building model with a square section. Vortex shedding effects were observed on the front corner surfaces resulting from corner modification, as with the side surface. For buildings with recessed corners, the local pressure on corner surfaces was larger than that of side surfaces. Moreover, the average wind pressure was effectively reduced to 88.42% and 92.40% in RE1 on the windward surface and CH1 on the side surface, respectively. | Tse, K. Family Income, Parenting Style, and Life Satisfaction in Parents and Adolescents | This study investigated the associations among family income and life satisfaction in parents, and parenting styles, income, and overall happiness and depression in their adolescents. A total of 1,913 participants comprising adolescents (aged 14-16) and their parents were recruited for this study. Participants were assessed using self-report questionnaires. Monthly household income was recorded by an open-ended question item. The life satisfaction of parents and overall happiness of adolescents were assessed by one item each, rated on a 4-point Likert scale. Three subtypes of parenting style (affectionate, monitoring, over-control) were evaluated by adolescents using an inventory consisting of 21 items on a 4-point Likert scale. Adolescents' depression was assessed with a checklist rated by the adolescents, consisting of 10 items on a 4-point Likert scale. The odds ratio (OR) for life dissatisfaction in adults was significantly higher in the lower (1st and 2nd) quintile groups of household income, by 9.94 (p < .001) and 3.50 (p < .001) folds, respectively, compared to the highest (5th) group. In contrast, household income had no significant association with life unhappiness in adolescents. Further, total household income had no significant association with the adolescent's depression score in linear regression analysis, but parenting style did. While affectionate (beta = -0.260; p < .001) and monitoring (beta = -0.100; p < .001) parenting styles showed negative associations with adolescents' depression scores, over-control style (beta = 0.120; p < .001) showed a positive association. | ○ | ○ | Article | Taxonomic Revision of the Leafhopper Genus Macropsis Lewis from Korea (Homoptera: Cicadellidae) | Korean species of the genus Macropsis Lewis are revised taxonomically, primarily based on the abdominal apodemes, male genitalia, and 2nd valvulae of the female ovipositor. Previously only three species were erroneously recorded from Korea. Among them, Macropsis notata (Prohaska, 1923) was previously misidentified as Macropsis prasina (Boheman, 1852). Two other European species, Macropsis scutellata (Boheman, 1845) and Macropsis illota (Horvath, 1899), are removed from the list of Korean fauna based on the examination of previously identified specimens that revealed only superficial resemblance to allied species. As a result, a total of 7 species are recognized in this revision, including 2 species new to science. Descriptions and distributional data are provided for all the species. | ○ | ○ | Article | The Most Important Aspects for a Good Death: Perspectives from Parents of Children with Cancer | A good death is an important concept in pediatric palliative care. To improve the quality of pediatric palliative care, it is imperative to identify which domain is most important for a good death among children with cancer and their parents. This study aimed to (1) assess the essential domains for a good death from the perspectives of parents whose children have cancer using the Good Death Inventory (GDI) and (2) examine which characteristics are associated with the perception of a good death. An anonymous cross-sectional questionnaire was administered to 109 parents of children with cancer. Data were collected using a validated Korean version of the GDI. Descriptive statistics, t-test, and ANOVA were used to identify the preferred GDI domains. Multiple linear regression analysis was performed to identify factors associated with the GDI scores. The most essential domains for a good death included "maintaining hope and pleasure" and "being respected as an individual." The factors most strongly associated with the perception of a good death were end-of-life plan discussion with parents or others and parental agreement with establishing a living will. | ○ | ○ | Article | Long-Term Effect of Silicone Oil Tamponade for Postoperative and Posttraumatic Bacterial Endophthalmitis | Purpose. To compare clinical features and microbial profiles, treatment outcomes, and prognostic factors of the eyes between postoperative and posttraumatic bacterial endophthalmitis after pars plana vitrectomy (PPV) with silicone oil (SO) tamponade. Methods. Overall, 57 eyes of 57 patients who diagnosed exogenous bacterial endophthalmitis and underwent PPV with SO tamponade between 2000 and 2019 were reviewed. Causative microorganisms, culture positivity, change of mean best-corrected visual acuity (BCVA), and course of treatment were investigated between postoperative and posttraumatic groups, and relevant factors were analyzed according to the final BCVA. Results. The mean BCVA change was not significantly different between groups. The positive rate of microorganisms was significantly higher in the postoperative group. The mean time to surgery over 48 hours, initial BCVA worse than hand motion, and additional surgery after initial vitrectomy were correlated with poor final BCVA worse than 20/200. There was significantly achieved final BCVA 20/200 or better in the Staphylococcus and Streptococcus group than the Enterococcus and Pseudomonas group. Conclusion. PPV with SO tamponade may be an effective surgical treatment strategy for exogenous bacterial endophthalmitis. | ○ | ○ | Article | Long-term Incidence of Kawasaki Disease in a North American Community: A Population-Based Study | To assess the longitudinal incidence of Kawasaki disease (KD) within the well-defined predominantly White population of Olmsted County, MN. This retrospective cohort study used a population-based medical record linkage system and manual chart reviews to identify children with KD in Olmsted County, MN between January 1, 1979-December 31, 2016. Age- and gender-adjusted incidence rates were calculated using the 2010 U.S. White population. 124 children with KD were confirmed during the study period (median age 3.5, 61% male, 85% White, 9% Asian). The overall age- and gender-adjusted incidence rates for all ages and < 5 years old were 9.8 and 21.4 per 100,000 person-years, respectively. There was an overall increase in incidence up to 1994 followed by plateau, except among children between the ages of 1-5 years. There was also an overall increase in incidence among females compared to males. 24% of children had cardiac complications. While the overall incidence of KD in Olmsted County appears to be stable since 1994, the incidence of KD in subgroups of children 1-5 years old and females seems to have increased.
