| ○ | ○ | Article | Spinal orexin A attenuates opioid-induced mechanical hypersensitivity in the rat | Background: Repeated administration of opioid analgesics for pain treatment can produce paradoxical hyperalgesia via peripheral and/or central mechanisms. Thus, this study investigated whether spinally (centrally) administered orexin A attenuates opioid-induced hyperalgesia (OIH). Methods: [D-Ala(2), N-Me-Phe(4), Gly-ol]-enkephalin (DAMGO), a selective mu-opioid receptor agonist, was used to induce mechanical hypersensitivity and was administered intradermally (4 times, 1-hour intervals) on the rat hind paw dorsum. To determine whether post- or pretreatments with spinal orexin A, dynorphin A, and anti-dynorphin A were effective in OIH, the drugs were injected through an intrathecal catheter whose tip was positioned dorsally at the L3 segment of the spinal cord (5 mu g for all). Mechanical hypersensitivity was assessed using von Frey monofilaments. Results: Repeated intradermal injections of DAMGO resulted in mechanical hypersensitivity in rats, lasting more than 8 days. Although the first intrathecal treatment of orexin A on the 6th day after DAMGO exposure did not show any significant effect on the mechanical threshold, the second (on the 8th day) significantly attenuated the DAMGO-induced mechanical hypersensitivity, which disappeared when the type 1 orexin receptor (OX1R) was blocked. However, intrathecal administration of dynorphin or an anti-dynorphin antibody (dynorphin antagonists) had no effect on DAMGO-induced hypersensitivity. Lastly, pretreatment with orexin A, dynorphin, or anti-dynorphin did not prevent DAMGO-induced mechanical hypersensitivity. Conclusions: Spinal orexin A attenuates mechanical hyperalgesia induced by repetitive intradermal injections of DAMGO through OX1R. | ○ | ○ | Article | Comparison of the efficacy and safety of LBAL, a candidate adalimumab biosimilar, and adalimumab reference product in patients with active rheumatoid arthritis inadequately responding to methotrexate: A 52-week phase III randomised study | Objective To evaluate the similarities between LBAL (adalimumab biosimilar candidate) and the adalimumab reference product (ADL) in terms of efficacy and safety, including immunogenicity, in patients with active rheumatoid arthritis despite methotrexate treatment. Methods This phase III, multicentre, randomised, double-blind, parallel-group, 56-week study was conducted in Japan and Korea. During the first 24 weeks, patients subcutaneously received 40 mg of LBAL or ADL every two weeks (LBAL and ADL groups). During the subsequent 28 weeks, the LBAL group patients and half of the ADL group patients received LBAL (L-L and A-L arms). The remaining ADL group patients continued to receive ADL (A-A arm). The primary efficacy endpoint was the change from baseline in disease activity score 28-erythrocyte sedimentation rate (DAS28-ESR) at Week 24. American College of Rheumatology (ACR) response rates, adverse events (AEs), and anti-drug antibody (ADA) were also assessed. Results In total, 383 patients were randomised. The least squares (LS) mean changes from baseline in DAS28-ESR at Week 24 were -2.45 and -2.53 in the LBAL (n=191) and ADL (n=190) groups, respectively. The 95% confidence interval (CI;-0.139, 0.304) of the difference (0.08) was within the pre-specified equivalence margin (-0.6, 0.6). Up to Week 52, the decreases in DAS28-ESR were maintained in all three arms. No notable differences in ACR20/50/70 were observed. The AE and ADA incidences were comparable between the arms. Conclusion LBAL was equivalent in efficacy and comparable in safety, including immunogenicity, to ADL. Switching from ADL to LBAL did not impact on efficacy and safety. | Matsuno, H.; Kang, Y. | ○ | ○ | Article | Implementation of a Geant4-based code using low-energy gamma-nuclear final state model for photonuclear studies | In this work, we developed a Geant4-based code, IS-2020, for photonuclear studies below 200 MeV covering the giant dipole resonance region (GDR). It uses G4LowEGammaNuclearModel, which consists of preequilibrium and equilibrium reaction mechanisms, for sampling the final states of photonuclear reactions. Simulations for photonuclear reactions on Mo-96, Zr-90, and the strongly-deformed nucleus Eu-153 targets have been performed for its validation. Moreover, a good agreement was also observed between experimental relative yields and the simulation results of Co-59(gamma,2n)Co-57 and Cr-50(gamma,2n)Cr-48 reactions induced by bremsstrahlung. The implemented code will be a useful and reliable tool for photonuclear studies, including designing the experimental setup, estimating the yield of outgoing products, and interpreting the measured data. | Anh, L. | ○ | ○ | Article | Measurement of the cluster position resolution of the Belle II Silicon Vertex Detector | The Silicon Vertex Detector (SVD), with its four double-sided silicon strip sensor layers, is one of the two vertex sub-detectors of Belle II operating at SuperKEKB collider (KEK, Japan). Since 2019 and the start of the data taking, the SVD has demonstrated a reliable and highly efficient operation, even running in an environment with harsh beam backgrounds that are induced by the world's highest instantaneous luminosity.In order to provide the best quality track reconstruction with an efficient pattern recognition and track fit, and to correctly propagate the uncertainty on the hit's position to the track parameters, it is crucial to precisely estimate the resolution of the cluster position measurement. | ○ | ○ | Article | Simulation of the Belle II silicon vertex detector | Belle II is the next generation B Factory experiment operating at the SuperKEKB accelerator complex at KEK in Tsukuba, Japan. It is expected to collect 50 ab(-1) of data, with a target instantaneous luminosity of 6.5 x 10(35) cm(-2)s(-1), which is about 30 times larger than its predecessor, Belle. In view of the ever increasing Belle II data sample, accurate simulation of the detector is growing in importance. This poses a challenging task of compromising between the realistic modeling of the response of individual detector components and reasonable performance in terms of CPU time of the simulation. | ○ | ○ | Article | Superiority of Simplified Acute Physiologic Score II Compared with Acute Physiologic and Chronic Health Evaluation II and Sequential Organ Failure Assessment Scores for Predicting 48-Hour Mortality in Patients Receiving Continuous Kidney Replacement Therapy | Background: Predicting early mortality is important in patients undergoing continuous kidney replacement therapy (CKRT), especially in the first 48 h. This study aimed to determine the predictive performance of the Simplified Acute Physiologic Score (SAPS) II, the Acute Physiologic and Chronic Health Evaluation (APACHE) II, and the Sequential Organ Failure Assessment (SOFA) scores for early mortality in patients receiving CKRT. Methods: Data from patients with acute kidney injury receiving CKRT were consecutively and retrospectively obtained at a tertiary medical center between August 2017 and March 2021. The outcomes included 48-h and 7-day mortality. The scoring systems were evaluated via discrimination at the time of CKRT initiation (using area under the receiver operating characteristics curve [AUROC]) and calibration (via Hosmer-Lemeshow goodness-of-fit C statistics). Results: Among eligible 652 patients, 95 (14.6%) and 212 (32.5%) died within 48 h and within 7 days, respectively. The AUROC for SAPS II (0.71, 95% confidence interval [CI]: 0.65-0.77, p = 0.016 vs. APACHE II score, p = 0.044 vs. SOFA score) was significantly higher than that of the APACHE II (0.66, 95% CI: 0.60-0.72) and SOFA scores (0.66, 95% CI: 0.60-0.72) for 48-h mortality. However, no significant differences in the AUROCs for SAPS II, APACHE II, and SOFA scores for 7-day mortality were observed. The calibration of the SAPS II for 48-h and 7-day mortality was adequate (p = 0.507 and p = 0.141, respectively). | ○ | ○ | Article | The Silicon Vertex Detector of the Belle II experiment | The Silicon Vertex Detector (SVD) is a part of the vertex detector in the Belle II experiment at the SuperKEKB collider (KEK, Japan). Since the start of data taking in spring 2019, the SVD has been operating stably and reliably with a high signal-to-noise ratio and hit efficiency, achieving good spatial resolution and high track reconstruction efficiency. The hit occupancy, which mostly comes from the beam-related background, is currently about 0.5% in the innermost layer, causing no impact on the SVD performance. In anticipation of the operation at higher luminosity in the following years, two strategies to sustain the tracking performance in future high beam background conditions have been developed and tested on data. One is to reduce the number of signal waveform samples to decrease dead time, data size, and occupancy. The other is to utilize the good hit-time resolution to reject the beam background hits. We also measured the radiation effects on the full depletion voltage, sensor current, and strip noise caused during the first two and a half years of operation. | ○ | ○ | Article | Determination of Volatility Parameters of Secondary Organic Aerosol Components via Thermal Analysis | To date, there are limited data on the thermal properties of secondary organic aerosol (SOA) components. In this study, we employed an experimental method to evaluate the physical properties of some atmospherically relevant compounds. We estimated the thermodynamic properties of SOA components, in particularly some carboxylic acids. The molar heat capacity, melting point and enthalpy, and vaporization enthalpy of the samples were determined via differential scanning calorimetry and thermogravimetric analysis, and their vaporization enthalpy (Delta H-vap) was estimated using Clausius-Clapeyron and Langmuir equations based on their thermogravimetric profiles. The thermodynamic properties of benzoic acid as a reference compound agree well with the reported values. The obtained specific heat capacities of benzoic acid, phthalic acid, pinic acid, ketopinic acid, cis-pinonic acid, terpenylic acid and diaterpenylic acid acetate (DTAA) are 118.1, 169.4, 189.9, 223.9, 246.1, 223.2, and 524.1 J mol(-1) K-1, respectively. The Delta H-vap of benzoic acid, phthalic acid, ketopinic acid, DTAA, and 3-methylbutane-1,2,3-tricarboxylic acid (3-MBTCA) are 93.2 +/- 0.4, 131.6, 113.8, and 124.4 kJ mol(-1), respectively. | ○ | ○ | Article | DN200434 Inhibits Vascular Smooth Muscle Cell Proliferation and Prevents Neointima Formation in Mice after Carotid Artery Ligation | Background: Excessive proliferation and migration of vascular smooth muscle cells (VSMCs), which contributes to the development of occlusive vascular diseases, requires elevated mitochondrial oxidative phosphorylation to meet the increased requirements for energy and anabolic precursors. Therefore, therapeutic strategies based on blockade of mitochondrial oxidative phosphorylation are considered promising for treatment of occlusive vascular diseases. Here, we investigated whether DN200434, an orally available estrogen receptor-related gamma inverse agonist, inhibits proliferation and migration of VSMCs and neointima formation by suppressing mitochondrial oxidative phosphorylation. Methods: VSMCs were isolated from the thoracic aortas of 4-week-old Sprague-Dawley rats. Oxidative phosphorylation and the cell cycle were analyzed in fetal bovine serum (FBS)- or platelet-derived growth factor (PDGF)-stimulated VSMCs using a Seahorse XF-24 analyzer and flow cytometry, respectively. A model of neointimal hyperplasia was generated by ligating the left common carotid artery in male C57BL/6J mice. Results: DN200434 inhibited mitochondrial respiration and mammalian target of rapamycin complex 1 activity and consequently suppressed FBS- or PDGF-stimulated proliferation and migration of VSMCs and cell cycle progression. Furthermore, DN200434 reduced carotid artery ligation-induced neointima formation in mice. | ○ | ○ | Article | Evaluation of Technology for the Analysis and Forecasting of Precipitation Using Cyclostationary EOF and Regression Method | Precipitation time series exhibit complex fluctuations and statistical changes. Existing research stops short of a simple and feasible model for precipitation forecasting. In this article, the authors investigate and forecast precipitation variations in South Korea from 1973 to 2021 using cyclostationary empirical orthogonal function (CSEOF) and regression methods. First, empirical orthogonal function (EOF) and CSEOF analyses are used to examine the periodic changes in the precipitation data. Then, the autoregressive integrated moving average (ARIMA) method is applied to the principal component (PC) time series derived from the EOF and CSEOF precipitation analyses. The fifteen leading EOF and CSEOF modes and their corresponding PC time series clearly reflect the spatial distribution and temporal evolution characteristics of the precipitation data. Based on the PC forecasts of the EOF and CSEOF models, the EOF-ARIMA composite model and CSEOF-ARIMA composite model are used to obtain quantitative precipitation forecasts. The comparison results show that both composite models have good performance and similar accuracy. However, the performance of the CSEOF-ARIMA model is better than that of the EOF-ARIMA model under various measurements. | ○ | ○ | Article | Metabolic Impacts of Discontinuation and Resumption of Recombinant Human Growth Hormone Treatment during the Transition Period in Patients with Childhood-Onset Growth Hormone Deficiency | Background: Discontinuing growth hormone (GH) treatment during the transition to adulthood has been associated with adverse health outcomes in patients with childhood-onset growth hormone deficiency (CO-GHD). This study investigated the metabolic changes associated with interrupting GH treatment in adolescents with CO-GHD during the transition period. Methods: This study included 187 patients with CO-GHD who were confirmed to have adult GHD and were treated at six academic centers in Korea. Data on clinical parameters, including anthropometric measurements, metabolic profiles, and bone mineral density (BMD) at the end of childhood GH treatment. were collected at the lime of re-evaluation for GHD and 1 year after treatment resumption. Results: Most patients (n=182, 97.3%) had organic GHD. The median age at treatment discontinuation and re-evaluation was 15.6 and 18.7 years, respectively. The median duration of treatment interruption was 2.8 years. During treatment discontinuation. body mass index Z-scores and total cholesterol. low-density lipoprotein. and non-high-density lipoprotein (HDL) cholesterol levels increased, whereas fasting glucose levels decreased. One year after GH treatment resumption, fasting glucose levels, HDL cholesterol levels, and femoral neck BMD increased significantly. Longer GH interruption (>2 years, 60.4%) resulted in worse lipid profiles at re-evaluation. The duration of interruption was positively correlated with fasting glucose and non-HDL cholesterol levels after adjusting for covariates. Conclusion: GH treatment interruption during the transition period resulted in worse metabolic parameters. and a longer interruption period was correlated with poorer outcomes. | ○ | ○ | Article | Regional characteristics of hot days and tropical nights in the Honam area, South Korea | This study analyses the regional characteristics of heat extremes (hot days and tropical nights) in the Honam area of South Korea during the recent 23 years (1997-2019) by using weather station data. The results based on the binary classifications of stations, that is, inland versus coastal and urban versus rural, show hot days in inland regions are climatologically more frequent, and present higher interannual variability and more evident increasing trend than coastal regions. On the other hand, inland regions are exposed to less active tropical nights. The thermal characteristics associated with the oceanic (or continental) climate at which the station is located are responsible for the inland-coastal dependency of frequencies in hot days and tropical nights. The noticeable discrimination between urban and rural stations, categorized by population, appears only in tropical night occurrences, while hot days are independent of urban-rural contrast. In urban stations, tropical night occurrences are climatologically higher, interannually more active, and recently more frequent. Regarding the urban heat island, the additional nocturnal heat leads to the urban concentrated activity of tropical nights.
