Article Enhanced primary ciliogenesis via mitochondrial oxidative stress activates AKT to prevent neurotoxicity in HSPA9/mortalin-depleted SH-SY5Y cells The primary cilium, an antenna-like structure on the cell surface, acts as a mechanical and chemical sensory organelle. Primary cilia play critical roles in sensing the extracellular environment to coordinate various developmental and homeostatic signaling pathways. Here, we showed that the depletion of heat shock protein family A member 9 (HSPA9)/mortalin stimulates primary ciliogenesis in SH-SY5Y cells. The downregulation of HSPA9 enhances mitochondrial stress by increasing mitochondrial fragmentation and mitochondrial reactive oxygen species (mtROS) generation. Notably, the inhibition of either mtROS production or mitochondrial fission significantly suppressed the increase in primary ciliogenesis in HSPA9-depleted cells. In addition, enhanced primary ciliogenesis contributed to cell survival by activating AKT in SH-SY5Y cells. The abrogation of ciliogenesis through the depletion of IFT88 potentiated neurotoxicity in HSPA9-knockdown cells. Furthermore, both caspase-3 activation and cell death were increased by MK-2206, an AKT inhibitor, in HSPA9-depleted cells. Taken together, our results suggest that enhanced primary ciliogenesis plays an important role in preventing neurotoxicity caused by the loss of HSPA9 in SH-SY5Y cells. Article Loosely synchronized activation of anterior cingulate cortical neurons for scratching response during histamine-induced itch Itch is a distinctive sensation that causes a specific affection and scratching reaction. The anterior cingulate cortex (ACC) has been linked to itch sensation in numerous studies; however, its precise function in processing pruritic inputs remains unknown. Distinguishing the precise role of the ACC in itch sensation can be challenging because of its capacity to conduct heterologous neurophysiological activities. Here, we used in vivo calcium imaging to examine how ACC neurons in free-moving mice react to pruritogenic histamine. In particular, we focused on how the activity of the ACC neurons varied before and after the scratching response. We discovered that although the change in neuronal activity was not synchronized with the scratching reaction, the overall activity of itch-responsive neurons promptly decreased after the scratching response. These findings suggest that the ACC does not directly elicit the feeling of itchiness. Article Nusinersen demonstrates effectiveness in treating spinal muscular atrophy: findings from a three-year nationwide study in Korea IntroductionNusinersen is the first drug approved for spinal muscular atrophy (SMA) treatment. In this study, we aimed to evaluate the long-term safety and efficacy of nusinersen, assess the therapeutic effects based on the treatment initiation timing and baseline motor function, and explore the perception of functional improvement from either parents or patients, utilizing 3-year nationwide follow-up data in South Korea.MethodsWe enrolled patients with SMA who were treated with nusinersen under the National Health Insurance coverage, with complete motor score records available and a minimum treatment duration of 6 months. To evaluate the motor function of patients, the Hammersmith Infant Neurological Examination-2 (HINE-2) was used for type 1 and the Expanded Hammersmith Functional Motor Scale (HFMSE) was used for types 2 and 3 patients. A significant improvement was defined as a HINE-2 score gain >= 5 for patients with type 1 and an HFMSE score >= 3 for patients with types 2 and 3 SMA. Effects of treatment timing were assessed. Patients with type 2 were further categorized based on baseline motor scores for outcome analysis. We also analyzed a second dataset from five tertiary hospitals with the information on parents/patients-reported impressions of improvement.ResultsThe study comprised 137 patients, with 21, 103, and 13 patients representing type 1, 2, and 3 SMA, respectively. At the 3-year follow-up, the analysis encompassed 7 patients with type 1, 12 patients with type 2, and none with type 3. Nearly half of all enrolled patients across SMA types (42.8, 59.2 and 46.2%, respectively) reached the 2-year follow-up for analysis. Patients with type 1 SMA exhibited gradual motor function improvement over 1-, 2-, and 3-year follow-ups (16, 9, and 7 patients, respectively). Patients with type 2 SMA demonstrated improvement over 1-, 2-, and 3-year follow-ups (96, 61 and 12 patients, respectively). Early treatment from symptom onset resulted in better outcomes for patients with type 1 and 2 SMA. In the second dataset, 90.7% of 108 patients reported subjective improvement at the 1-year follow-up.ConclusionNusinersen treatment for types 1-3 SMA is safe and effective in long-term follow-up. Early treatment initiation was a significant factor affecting long-term motor outcome. Review Role of spinal astrocytes through the perisynaptic astrocytic process in pathological pain Pathological pain is caused by abnormal activity in the neural circuit that transmits nociceptive stimuli. Beyond homeostatic functions, astrocytes actively participate in regulating synaptic transmission as members of tripartite synapses. The perisynaptic astrocytic process (PAP) is the key structure that allows astrocytes to play these roles and not only physically supports synapse formation through cell adhesion molecules (CAMs) but also regulates the efficiency of chemical signaling. Accumulating evidence has revealed that spinal astrocytes are involved in pathological pain by modulating the efficacy of neurotransmitters such as glutamate and GABA through transporters located in the PAP and by directly regulating synaptic transmission through various gliotransmitters. Although various CAMs contribute to pathological pain, insufficient evidence is available as to whether astrocytic CAMs also have this role. Therefore, more in-depth research is needed on how pathological pain is induced and maintained by astrocytes, especially in the PAP surrounding the synapse, and this will subsequently increase our understanding and treatment of pathological pain. Article NO2 Gas Sensing Properties of Ag-Functionalized Porous ZnO Sheets Herein, we report a simple and scalable synthesis route to prepare Ag-functionalized porous ZnO sheets and their enhanced NO2 gas sensing properties. Porous ZnO sheets functionalized with well-dispersed submicron Ag particles were prepared by using a hydrothermal method-based one-pot synthesis route from Zn and Ag precursors. NO2 gas sensing performance (response, selectivity, response time, and recovery time) was optimized at 200 degrees C in the gas sensor fabricated with 3 at% Ag-functionalized porous ZnO sheets. We demonstrated a response (R-g/R-a) of 17.18 to 10 ppm NO2 gas and also obtained a high response of 14.05 even at 60% relative humidity due to the synergetic effect of improved NO2 gas adsorption in the presence of Ag particles and increased resistance by the formation of Schottky barrier at Ag-ZnO heterojunctions. Article Applying multi-focus filtering plane in double-step Fresnel diffraction method for enhanced holographic projection region Computer-generated holography has been anticipated in augmented reality (AR) since it can fully provide multi-depth 3D information to users, but speckle noises are often accompanied as one of the most significant degradation factors. Since complex amplitude encoding can suppress speckle noises, applying double-phase encoding to a lensless holographic projection system using a double-step Fresnel diffraction (DSF) algorithm has been reported to reduce those noises. However, a single filtering focus at a virtual focal plane might significantly reduce the projection region compared to the maximum diffraction bandwidth of the spatial light modulator. In this paper, we applied multiple foci for the focal plane of the DSF method with off-axis phase encoding, which can expand the projection region and is applicable to display multi-depth images with DSF. Numerical analysis conducted for image quality compared with conventional single focus DSF algorithm shows that the speckle performance does not degrade so much as the number of foci increase. We expect the proposed method to be useful for holographic projection optical systems such as AR head-up display. Article Bulk Bi2Te3-based bendable thermoelectric device with highly elastic Cu-Be alloy foils The binary Cu-Be alloy features high elasticity and yield strength. Hence, it can be used as an electrode material for bulk Bi2Te3-based thermoelectric (TE) devices to create bendability. First, TE materials and Cu-based electrodes were both coated with Ni/Au multilayers, which acted as diffusion barriers. Subsequently, the TE legs were connected to the Cu alloy electrode via soldering. Diffusion barriers enhanced the bonding strength and decreased the internal resistance of the TE generator. Furthermore, the internal resistance remained stable after a cyclic bending test, indicating the high flexibility of the device. The cooling performance was estimated by measuring the temperature on one side of the device while various current intensities were applied. The output power was measured using different temperature gradients and compared to the output power generated by a thermoelectric device for heat treatment. Article Characteristics of phosphorescent organic light-emitting devices with interfacial nanolayers of acid molecule-doped tris(4-aminophenyl)amine Here we report that 2-acrylamido-2-methyl-1-propanesulfonic acid (AMPSA)-doped tris(4-aminophenyl)amine (TAPA) can act as an interfacial layer for hole injection in phosphorescent organic light-emitting devices (OLEDs) and deliver improved device stability upon electric fields. The acid-base reaction between TAPA and AMPSA was carried out using 2-methoxy ethanol solvent with various molar ratios (0-300 mol. %). The resulting TAPA:AMPSA layers (1-10 nm thick) were spin-coated on indium-tin-oxide (ITO)-glasses and selectively annealed before the thermal deposition of organic small molecular multilayers in a vacuum. The doping reaction between TAPA and AMPSA gave rise to a new absorption peak at a wavelength of ca. 880 nm. The work function of TAPA:AMPSA layer-coated ITO-glasses increased with the molar ratio, but the highest luminance and efficiency of OLEDs were achieved at 100 mol. % (AMPSA to TAPA). The thickness control and thermal treatment process of TAPA:AMPSA layers (100 mol. %) could further enhance the luminance (from ca. 5000 cd/m(2) (t = 10 nm) to 15,500 cd/m(2) (t = 2 nm)) with relatively high device stability upon electric field compared to the pristine TAPA layers. Article High-performance tin oxide field-effect transistors deposited by thermal atomic layer deposition In this study, we investigated the impact of annealing ambient on the performance of SnO2-based field-effect transistors (FETs) while adhering to the thermal budget constraint (process temperature < 450 degree celsius). The SnO2 films were deposited by thermal atomic layer deposition (T-ALD). After annealing the films for 2 h at 400 degree celsius, it was observed that annealing in an O-2 ambient resulted in a SnO2 film with an excessive number of background carriers, which made it difficult to fully deplete the channel. Conversely, annealing in an N-2 ambient led to an unstable channel that could not be effectively modulated, despite having a carrier concentration about an order of magnitude lower than that of the O-2-annealed film. Surprisingly, the devices annealed in an air ambient exhibited the best performance. X-ray photo-emission spectroscopy revealed that annealing in O-2 ambient resulted in a higher level of oxygen vacancy, while N-2 annealing was more effective in creating stoichiometric SnO. The best device performance achieved in this study was observed in a 6-nm thick channel that was annealed in the air. This device exhibited an on/off ratio of 1.2 x 10(11), subthreshold swing of 0.4 V/dec., field-effect mobility of 14.6 cm(2)/V center dot s, and threshold voltage of 1.1 V, operating in an enhancement mode. These results emphasize the potential of ALD SnO2 channels for high-performance transistors that are compatible with back-end-of-line processes in Si technology.
