| ○ | ○ | Review | The π-π stacking effect of metallocene catalyst in olefin polymerization | The single-sited metallocene catalyst has attracted significant attention due to its high activity and application in olefin polymerization industry. In this work, we introduced a strategy of in situ modification of metallocene catalyst with pyrene through pi-pi stacking to study the effect in olefin polymerization. After activated by MMAO, the pyrene-modified metallocene catalyst exhibited high activities (up to 1333 kg mol(-1) h(-1)) in ethylene polymerization, which was 1.9 times higher than that of the pyrene free catalyst system. This could be because the electron-donating ability of cyclopentene to the transition metal was enhanced through pi-pi interaction of pyrene with cyclopentene. The copolymerization of ethylene and 1-hexene was also studied. The pyrene-modified metallocene catalyst system performs with higher activity in the copolymerization with a lower monomer incorporation ratio. | ○ | ○ | Article | A Novel Method of Self-Cross-Linking of Syringaldehyde with Activated Methoxy Groups via Cross-Coupling for Lignin-Based Wood Adhesives | As steric hindrance, methoxy groups are limiting the valorization of hardwood lignin. This paper reports a novel method of self-cross-linking of the syringaldehyde with activated methoxy groups (-OCH3) via cross-coupling reaction to obtain thermosetting polymers for lignin-based wood adhesives. The methoxy groups of syringaldehyde have been activated via cross-coupling reaction by substituting Ar-OCH3 with Ar-CH2-SiMe3, and dichloromethane, leading to cross-linking via methylene bridges to build a thermosetting polymer. FTIR spectra showed a decrease in the intensity of a -CH3 and -OH group, owing to the substitution of the methoxy group. C-13 NMR spectra also supported these results with the -SiMe3 signal that disappeared after the cross-linking reaction. Furthermore, cross-linking between the activated methoxy groups was confirmed with a strong exothermic peak at 130 degrees C, resulting in an increase in the adhesion strength as hot-pressing temperature increased from 160 to 180 degrees C. | ○ | ○ | Article | Engineering Three-Dimensional Spheroid Culture for Enrichment of Proangiogenic miRNAs in Umbilical Cord Mesenchymal Stem Cells and Promotion of Angiogenesis | In the field of regenerative medicine, umbilical cord-derived mesenchymal stem cells (UC-MSCs) have a plausible potential. However, traditional two-dimensional (2D) culture systems remain limited in replicating the complex in vivo microenvironment. Thus, three-dimensional (3D) cultures offer a more physiologically relevant model. This study explored the impact of 3D culture conditions on the UC-MSC secretome and its ability to promote angiogenesis, both in vitro and in vivo. In this study, using two distinct methods, we successfully cultured UC-MSCs: in a monolayer (2D-UC-MSCs) and as spheroids formed in U-shaped 96-well plates (3D-UC-MSCs). The presence and expression of proangiogenic miRNAs in the conditioned media (CM) of these cultures were investigated, and differential expression patterns were explored. Particularly, the CM of 3D-UC-MSCs revealed significantly higher levels of miR-21-5p, miR-126-5p, and miR-130a-3p compared to 2D-UC-MSCs. Moreover, the CM from 3D-UC-MSCs revealed a higher effect on endothelial cell proliferation, migration, and tube formation than did the CM from 2D-UC-MSCs, indicating their proangiogenic potential. | ○ | Article | Experimental Investigation of Electrospraying Properties Based on Ring Electrode Modification | Electrospraying uses a high-voltage potential difference to create fine droplets. This study conducts a comparative analysis of the spray pattern and droplet properties using ring electrode parameters. The spray pattern and droplet characteristics are analyzed based on the experimental parameters of the ring electrode. The results show that the cone-jet mode forms quickly for the ring electrode. In addition, as the ring diameter decreases, the ring voltage increases and an increase in the distance between the ring and the nozzle in the bottom direction decreases the Sauter mean diameter and its standard deviation. The optimal conditions for the formation of fine and uniform droplets include a ring diameter of 15 mm, a ring voltage of 7 kV, and a nozzle-to-ring distance of (+) 20 mm. © 2023 The Authors. | ○ | ○ | Article | Exploring the Anti-inflammatory Potential of Novel Chrysin Derivatives through Cyclooxygenase-2 Inhibition | Inducible cyclooxygenase-2 (COX-2) is a crucial enzyme involved in the processes of inflammation and carcinogenesis, primarily by catalyzing the production of prostaglandin E2 (PGE2), a significant mediator of inflammation. In this study, we designed and synthesized a series of novel chrysin derivatives to evaluate their anti-inflammatory potential through COX-2 inhibition using in vitro cultures of RAW264.7 macrophages and in silico molecular docking assays. Among the synthesized derivatives, compounds 1a and 8 demonstrated significant inhibition of lipopolysaccharide (LPS)-stimulated proinflammatory cytokine production, including interleukin-6 and tumor necrosis factor-alpha, in RAW264.7 cells. Additionally, these derivatives effectively inhibited PGE2 secretion through COX-2 enzyme inhibition in LPS-stimulated RAW264.7 cells. Molecular docking simulation results revealed that 1a and 8 possess high binding affinities for the COX-2 active site, indicating a strong potential for enzyme inhibition. Furthermore, druglikeness and ADMET predictions for these compounds indicated favorable pharmacokinetic properties, suggesting their suitability as drug candidates. | ○ | ○ | Article | Fungal Microbiome of Alive and Dead Korean Fir in its Native Habitats | A rapid decline of Abies koreana has been reported in most of the natural alpine habitats in Korea. It is generally accepted that this phenomenon is due to climate change even though no clear conclusions have been drawn. Most research has focused on abiotic environmental factors, but studies on the relationships between A. koreana and soil fungal microbiomes are scarce. In this study, the rhizoplane and rhizosphere fungal communities in the alive and dead Korean fir trees from its three major natural habitats including Mt. Deogyu, Mt. Halla, and Mt. Jiri in Korea were investigated to identify specific soil fungal groups closely associated with A. koreana. Soil fungal diversity in each study site was significantly different from another based on the beta diversity calculations. Heat tree analysis at the genus level showed that Clavulina, Beauveria, and Tomentella were most abundant in the healthy trees probably by forming ectomycorrhizae with Korean fir growth and controlling pests and diseases. However, Calocera, Dacrymyces, Gyoerffyella, Hydnotrya, Microdochium, Hyaloscypha, Mycosymbioces, and Podospora were abundant in the dead trees. Our findings suggested that Clavulina, Beauveria, and Tomentella are the major players that could be considered in future reforestation programs to establish ectomycorrhizal networks and promote growth. These genera may have played a significant role in the survival and growth of A. koreana in its natural habitats. In particular, the genus Gyoerffyella may account for the death of the seedlings. | ○ | ○ | Article | Glucaric Acid Production from Miscanthus sacchariflorus via TEMPO-Mediated Oxidation with an Efficient Separation System | In this study, production and isolation of glucaric acid from lignocellulosic biomass were performed via potassium cation-based TEMPO-mediated oxidation for the ease of glucaric acid isolation. To optimize the oxidation conditions, response surface methodology (RSM) was adopted using standard glucose as the raw material. Among the oxidation conditions, the dosage of oxidant and pH of reaction affected the glucaric acid production, and the optimum conditions were suggested by RSM analysis: 5 C-degrees of reaction temperature, 4.23 equiv dosage of KClO per mole of glucose, and pH of 12. Furthermore, glucaric acid was produced from lignocellulosic biomass-derived enzymatic hydrolysate from Miscanthus under optimum conditions. The impurities such as xylose and lignin in enzymatic hydrolysate inhibited the efficiency of glucose oxidation. As a result, more oxidant was required to produce sufficient glucaric acid from the enzymatic hydrolysate compared to standard glucose. The produced glucaric acid was simply isolated by controlling the pH in the form of glucaric acid monopotassium salt, which showed lower solubility in water, and the purity of isolated glucaric acid was over 99%. | ○ | ○ | Article | Molecular Identification of a New Species of Absidia (Cunninghamellaceae, Mucorales) Isolated from Soil in Korea | The species within the family Cunninghamellaceae during an investigation of soil microfungi in Korea, in which three strains were isolated from Gangwon, Chungbuk, and Gyeongbuk provinces, designated as KNUF-22-121A, KNUF-22-126A, and KNUF-22-316, respectively. Because the morphological and molecular analyses of these three strains were identical, KNUF-22-316 underwent further detailed study. Phylogenetic analyses based on the concatenated nucleotide sequences of the internal transcribed spacer region and the large subunit 28S rRNA gene revealed that the strain belonged to the genus Absidia, but occupied a distinct phylogenetic position. The strain KNUF-22-316 was compared with closely related species Absidia radiata CGMCC 3.16257(T) and Absidia yunnanensis CGMCC 3.16259(T), morphologically different with shorter sporangiophores, smaller sporangia and columellae, and the consistent presence of collars. | ○ | ○ | Article | Morphological and Phylogenetic Characterization of Trichomerium koreanum sp. nov. and Exophiala radicis Belonging to Chaetothyriales Fungi in Korea | The order Chaetothyriales encompasses black yeasts and related fungi known for their diverse morphologies, asexual reproduction, and habitat preferences. In this study, we isolated two fungal strains belonging to Chaetothyriales, designated KNUF-23-13A and KNUF-22-020, from a longhorn beetle (Cyrtoclytus capra) and soil sample, respectively, in Korea. Phylogenetic analyses based on the concatenated nucleotide sequences of the internal transcribed spacer regions and partial sequences of the large subunit (LSU) rRNA, beta-tubulin (TUB2), and actin (ACT) genes revealed that the strains were grouped with strains of Trichomerium and Exophiala, respectively. For strain KNUF-23-13A, cultural and morphological differences were observed in the shape and size of mycelium between it and its phylogenetically closest species, Trichomerium lapideum CGMCC 3.17311(T), supporting the novelty of this isolated species. In the case of strain KNUF-22-020, the cultural and morphological characteristics were similar to those previously reported for E. radicis P2854T. | ○ | ○ | Article | Phylogenetic and Morphological Analysis of a Novel Tolypocladium Species Isolated from Soil in South Korea | A fungal strain designated KNUF-22-014, belonging to the family Ophiocordycipitaceae, was isolated from a soil sample collected in Sobaeksan National Park, South Korea. Phylogenetic trees based on the combined sequences of internal transcribed spacer (ITS) regions and the large subunit of 28S rRNA (LSU) gene, as well as on five concatenated loci (LSU, ITS, translation elongation factor 1 alpha, beta-tubulin, and subunit 1 of RNA polymerase II), revealed that the isolate falls within the clade containing the genus Tolypocladium but occupies a distinct phylogenetic position. Its closest relative is T. tropicale. Morphologically, strain KNUF-22-014 can be distinguished from T. tropicale by the shape of its phialides (subcylindrical vs. lageniform), its conidiophore type (arborescent or unbranched vs. trichodermatoid), and its larger conidia size (2.1 +/- 0.1 mu m vs. 1.5 +/- 0.1 mu m). Nov.: A Novel Fungal Species Belonging to the Family Podosporaceae Isolated from Soil in Korea | The fungal strain designated as KNUF-21-020, belonging to the genus Triangularia, was isolated from a soil sample collected in the Chungnam province, Korea. Phylogenetic analyses based on the concatenated nucleotide sequences of internal transcribed spacer regions and partial sequences of large subunit rRNA, beta-tubulin, and RNA polymerase II subunit genes revealed that the strain was grouped in a clade with Triangularia species. However, it occupied a distinct phylogenetic position. We also observed morphological differences between strain KNUF-21-020 and closely related species. Here, we provided detailed descriptions, illustrations, and discussions regarding the morphological and phylogenetic analyses of the closely related species to support the novelty of this isolated species. The phylogenetic analyses and morphological observations indicate that the strain KNUF-21-020 represents a novel species in the genus Triangularia (family: Podosporaceae). | ○ | ○ | Article | Atypically located accessory splenic hemangiosarcoma in a Dog: Diagnostic value of triple-phase computed tomography | A 10-year-old Cocker spaniel presented with lethargy. Triple-phase computed tomography was obtained with a contrast test bolus at the level of porta hepatis, which revealed a right lower abdominal mass. The mass was not connected to other abdominal organs; however, a linear structure was observed connecting the splenic hilum to the mass, which was suspected to be the feeding vessel. The arterial phase image was obtained again with a contrast bolus at the level of the celiac artery. A prominent contrast-enhanced feeding artery originating from the splenic artery to the mass was observed. Histopathology confirmed an accessory splenic hemangiosarcoma. Graphical Abstract: In a 10-year-old Cocker spaniel, an ultrasound revealed a right caudal abdominal mass. On triple-phase CT, the mass was not connected to adjacent organs, but feeding vessels from the spleen connecting to the right caudal abdominal mass were observed. | ○ | ○ | Article | Effect of the sodium-glucose cotransporter-2 inhibitor, DWP16001, as an add-on therapy to insulin for diabetic dogs: A pilot study | Background: Sodium-glucose cotransporter-2 (SGLT2) inhibitors are a novel class of anti-hyperglycaemic agents. Objective: This study aimed to evaluate the safety and the adjuvant glycaemic control effect of an SGLT2 inhibitor, DWP16001, in diabetic dogs receiving insulin treatment. Methods: Nineteen diabetic dogs receiving insulin treatment (NPH, porcine lente and glargine insulin) were divided into two groups according to dosing frequency: DWP TOD group (n = 10) and DWP SID group (n = 9). In the DWP TOD group, 0.025 mg/kg of DWP16001 was administered once every 3 days, whereas, in the DWP SID group, 0.025 mg/kg of DWP16001 was administered once a day. Food intake was maintained during the trial period. Hypoglycaemia, ketoacidosis or unexpected life-threatening reactions were assessed as adverse effects before and after DWP16001 administration. We compared insulin requirement reduction and blood glucose level control between two groups. Results: No specific adverse effects were observed during the clinical trial, and haematological parameter remained unchanged. Moreover, the fasting glucose levels and daily insulin dose in the DWP TOD group were lower than the pre-administration values, but not significantly different for 8 weeks. Systolic blood pressure, fructosamine and insulin dose decreased significantly in the DWP SID group compared to the DWP TOD group at 8 weeks (p < 0.05) without affecting food consumption. Among these patients, 10 patients were monitored while receiving DWP16001 for 12 months (DWP TOD group n = 5, DWP SID group n = 5). The fasting glucose and fructosamine levels and daily insulin dose were reduced in both groups at 12 months compared with those before receiving DWP16001. | ○ | ○ | Article | Evaluation of haematological parameters in haemolytic anaemia caused by tick-borne pathogens in grazing cattle | BackgroundNo tick-borne pathogens (TBPs) causing haemolytic anaemia in cattle have been reported, except Theileria orientalis and complete blood count (CBC) profile is the only haematological parameter to determine the severity of regenerative haemolytic anaemia.ObjectivesTo identify the causative agents of TBP-induced haemolytic anaemia and determine haematological parameters that indicate haemolytic anaemia in grazing cattle.MethodsEighty-two Korean indigenous cattle (Hanwoo) were divided into two groups: grazing (n = 67) and indoor (n = 15) groups. CBC and serum biochemistry were performed. PCR was conducted using whole blood-extracted DNA to investigate the prevalence of TBPs.ResultsTBP-induced haemolytic anaemia was observed in the grazing group. In grazing cattle, co-infection (43.3%, 29/67) was most frequently detected, followed by T. orientalis (37.6%, 25/67) and Anaplasma phagocytophilum infections (1.5%, 1/67). In indoor cattle, only co-infection (20%, 3/15) was identified. Grazing cattle exhibited regenerative haemolytic anaemia with marked monocytosis, mild neutropenia, and thrombocytopenia. According to grazing frequency, the 1st-time grazing group had more severe anaemia than the 2nd-time grazing group. Elevations in indirect bilirubin and L-lactate due to haemolytic anaemia were identified, and correlations with the respective markers were determined in co-infected grazing cattle.ConclusionsQuantitative evaluation of haematocrit, mean corpuscular volume, and reticulocytes (markers of regenerative haemolytic anaemia in cattle) was performed for the first time. Our results show that, in addition to T. orientalis, A. phagocytophilum is strongly associated with anaemia. The correlation between haemolytic anaemia severity and haematological parameters (indirect bilirubin, reticulocytes, and L-lactate) was confirmed.
