| ○ | ○ | Article | Beyond CHD7 gene: unveiling genetic diversity in clinically suspected CHARGE syndrome | The Verloes or Hale diagnostic criteria have been applied for diagnosing CHARGE syndrome in suspected patients. This study was conducted to evaluate the diagnostic rate of CHD7 according to these diagnostic criteria in suspected patients and also to investigate other genetic defects in CHD7-negative patients. The clinical findings and the results of genetic testing of CHD7, chromosome microarray, exome sequencing, or genome sequencing of 59 subjects were reviewed. CHD7 pathogenic variants were identified in 78% of 46 subjects who met either the Verloes or Hale diagnostic criteria and in 87% of 38 subjects who met both criteria, whereas no CHD7 variant was detected in 13 subjects who met neither criterion. Among 23 patients without the CHD7 variant, six genetic diseases were identified in 7 patients, including Wolf-Hirschhorn syndrome, 1q21 deletion syndrome, 19q13 microdeletion, and pathogenic variants in PLCB4, TRRAP, and OTX2. Based on these comprehensive analyses, the overall diagnostic rate was 73% for seven different genetic diseases. NIBRBAC000502770 from coal mine of Hongcheon on Republic of Korea | ObjectivesThe data were collected to obtain the complete genome sequence of Pseudarthrobacter sp. NIBRBAC000502770, isolated from the rhizosphere of Sasamorpha in a heavy metal-contaminated coal mine in Hongcheon, Republic of Korea. The objective was to explore the strain's genetic potential for plant growth promotion and heavy metal resistance, particularly arsenate and copper. The aim focused on identifying microbes that enhance plant growth in metal-tolerant environments and evaluating the strain's bioremediation and agricultural uses. This study sought key genes for bioremediation and agricultural applications in contaminated soils, aiding sustainable management and biotechnology.Data descriptionWe report the complete genome sequence of Pseudarthrobacter sp. NIBRBAC000502770, isolated from a coal mine in Hongcheon, Republic of Korea. The genome contains a chromosome (4,403,796 bp) and a plasmid (74,326 bp, named pMK-1) with 286-fold coverage. Genome annotation identified 4,209 genes, including 3,926 protein-coding genes, 51 tRNA genes, and 15 rRNA genes, with a G + C content of 66.1%. Functional analysis revealed genes related to plant growth promotion and heavy metal resistance, such as arsenate (arsR, arsC) and copper (copC, copD) resistance genes. Genes involved in auxin biosynthesis suggest potential agricultural applications. | ○ | ○ | Article | Effects of PM2.5 Metal Components Derived From Porcine Farm Exposure on Sperm Function in Mice | This study aimed to identify the effects of major metal components present in particulate matter (PM)2.5 on the reproductive system, sperm function, and embryo development. Through intratracheal instillation, male mice were exposed to various concentrations of metal components, including calcium oxide (Ca), iron oxide (Fe), aluminum oxide (Al), zinc oxide (Zn), lead oxide (Pb), and a mixture of these metals, in PM2.5 collected from the porcine farm. After 14 days, testicular inflammation and abnormal sperm morphology were observed in the exposed mice. These results indicate that such metal exposure enhances inflammatory cytokines in the testis and oxidative stress-induced apoptosis. Moreover, the exposure influenced sperm deformation, capacitation status, testosterone levels, and testosterone biosynthesis. Importantly, embryo development was also found to be impacted due to decreased sperm fertility. | ○ | ○ | Article | Investigation of the Space Charge Dynamic in the Nanocomposite BaTiO3 -Doped XLPE | Understanding space charge dynamics in cross-linked polyethylene (XLPE) is crucial for optimizing electrical insulation systems, preventing insulation failure, and ensuring the reliable performance of electrical equipment. Research is focused on advanced characterization techniques and predictive models to mitigate space charge effects, which are key factors in the aging and degradation of high-voltage polymer insulators. While experimental measurements are common, theoretical models are less so. This paper investigates the dynamics of space charges in barium titanate (BaTiO3)-doped XLPE nanocomposites under a continuous electric field using the bipolar charge transport model. The results show that doping with BaTiO3 significantly improves the dielectric properties of XLPE. | ○ | ○ | Article | Microiontophoretic Application of Dynorphin in Dental Pain: Excitatory or Inhibitory Effects | Background: The tooth exhibits increased sensitivity to noxious stimuli due to the dense innervation of thin myelinated A delta fibers and unmyelinated C fibers within the dental pulp. While prior research has identified dynorphin expression in layers I-II of the dorsal horn across the spinal cord in various pain models, its functional role in trigeminal nociception, including tooth pain, remains under- explored. This study examines the potential role of dynorphin in the nociceptive processing of dental stimuli. Methods: Experiments were performed on adult male ferrets weighing 0.9-1.4 kg. The effects of dynorphin on electrically evoked responses of tooth pulp neurons were recorded extracellularly. Results: The results demonstrated that the microiontophoretic application of dynorphin A induced excitatory and inhibitory effects on N-methyl-D-aspartate (NMDA)-evoked responses in electrically stimulated tooth pulp neurons. Specifically, dynorphin A attenuated NMDA-evoked responses in 16 out of 32 neurons by 61 +/- 6%, facilitated NMDA-evoked responses in 10 out of 32 neurons by 69 +/- 17%, and elicited mixed inhibitory and facilitatory responses in six out of 32 neurons. The inhibitory effects of dynorphin were blocked by nor-binaltorphimine, a kappa receptor antagonist, whereas the facilitatory effects were inhibited by D,L-2-amino5-phosphonovaleric acid, an NMDA receptor antagonist. Conclusion: These findings suggest that dynorphin A-induced excitatory responses are mediated by NMDA receptors, whereas its inhibitory responses are mediated through kappa opioid receptors in dental pain. | ○ | ○ | Article | Impact of preoperative prognostic factors on survival outcomes in intrahepatic cholangiocarcinoma: a retrospective cohort study | Purpose: This study assesses preoperative prognostic factors in intrahepatic cholangiocarcinoma (ICC) to improve risk assessment and inform clinical decisions, focusing on the role of lymph node dissection (LND). Methods: We conducted a retrospective analysis of 234 patients who underwent surgery for ICC at a single institution between 2010 and 2019. Prognostic factors affecting overall survival (OS) were identified through univariate and multivariable logistic regression analyses. Patients were categorized into high-, intermediate-, and low-risk groups based on the number of adverse prognostic factors. Survival curves were compared between the groups that underwent LND and those that did not within each risk category. Results: Of the 234 patients, 138 (59.0%) underwent LND, and 96 (41.0%) did not (non-LND). Significant prognostic factors included preoperative elevation of CEA (>5 ng/mL), CA 19-9 (>37 U/mL), tumor multiplicity, tumor size >5 cm, and extrahepatic invasion. These factors were associated with adverse OS (hazard ratio ranging from 1.69 to 2.54). High-risk patients had significantly lower median OS compared to intermediate and low-risk groups in both LND and non-LND cohorts, but no significant difference in median OS between LND and non-LND groups within each risk category was observed. Conclusion: Preoperative prognostic factors such as CEA, CA 19-9, tumor size, and multiplicity are vital for assessing patient risk in ICC. These factors guide clinical decision-making and emphasize the need fortargeted treatment strategies, including the consideration of LND, particularly in high-risk patients. | ○ | ○ | Article | Postoperative results after open-wedge high tibial osteotomy with lateral hinge fracture | Background: Lateral hinge fracture (LHF) is a known complication of open-wedge high tibial osteotomy (OWHTO), potentially resulting in postoperative instability, displacement, loss of correction, malunion, and non-union. This study tested the hypothesis that patients who develop LHF after OWHTO can achieve clinical outcomes comparable to those without LHF, without requiring additional surgical intervention. Methods: This retrospective study included 96 patients who underwent OWHTO with a locking plate between 2019 and 2021. Postoperative radiographs and computed tomography (CT) scans identified LHF in 24 patients (25%). Lower limb alignment was assessed using whole-leg standing radiographs at 6 weeks, 3 months, 6 months, and 12 months postoperatively. Clinical outcomes were evaluated using the Hospital for Special Surgery (HSS) score, Knee Society Knee Score (KS), and Function Score (FS). Results: In patients with LHF, the mechanical axis was 59.0 +/- 7.4% at 6 weeks, 57.7 +/- 7.8% at 3 months, 55.9 +/- 9.5% at 6 months, and 53.9 +/- 12.2% at 12 months. In patients without LHF, the corresponding values were 58.5 +/- 6.1%, 57.8 +/- 6.7%, 56.7 +/- 7.0%, and 55.6 +/- 7.4%, respectively. Although the mechanical axis decreased by 5.2% in the LHF group and 2.9% in the non-LHF group by 12 months, the difference was not statistically significant (p = .219). Similarly, the longitudinal change in alignment did not differ significantly between groups (p = .743). Postoperative clinical scores improved in all patients, with no significant differences between groups regardless of LHF status. | ○ | ○ | Article | Exploration of silk-hyaluronic acid hydrogels crosslinked by the Fenton reaction | Herein, HRP-free silk-HA hybrid hydrogels are generated using the Fenton reaction. The findings reveal that a higher HA concentration, at an appropriate crosslinking density, increases the random coil content while decreasing the beta-sheet content by reducing the interactions of Fe3+ ions with silk fibroin. This facilitates the formation of homogeneous hydrogel networks with enhanced optical transparency. | ○ | ○ | Review | Is platelet-rich plasma better than steroids as epidural drug of choice in lumbar disc disease with radiculopathy? Meta-analysis of randomized controlled trials | The current meta-analysis was performed to analyze the efficacy and safety of platelet-rich plasma (PRP) as an epidural injectate, in comparison with steroids in the management of radiculopathy due to lumbar disc disease (LDD). We conducted independent and duplicate searches of the electronic databases (PubMed, Embase and Cochrane Library) in March 2024 to identify randomized controlled trials (RCTs) analyzing the efficacy of epidural PRP for pain relief in the management of LDD. Animal or in vitro studies, clinical studies without a comparator group, and retrospective or non-randomised clinical studies were excluded. Diverse post-intervention pain scores [visual analog score (VAS)] and functional scores [Oswestry Disability Index (ODI), SF-36], as reported in the reviewed studies, were evaluated. Statistical analysis was performed using STATA 17 software. 5 RCTs including 310 patients (PRP/Steroids = 153/157) were included in the analysis. The included studies compared the efficacy and safety of epidural PRP and steroids at various time-points including 1, 3, 6, 12, 24, and 48 weeks. Epidural PRP injection was found to offer comparable pain relief (VAS; WMD = -0.09, 95% CI [-0.66, 0.47], p = 0.641; I-2 = 96.72%, p < 0.001), functional improvement (ODI; WMD = 0.72, 95% CI [-6.81, 8.25], p = 0.524; I-2 = 98.73%, p < 0.001), and overall health improvement (SF-36; WMD = 1.01, 95% CI [-1.14, 3.17], p = 0.224; I-2 = 0.0%, p = 0.36) as epidural steroid injection (ESI) at all the observed time points in the included studies without any increase in adverse events or complications. Epidural administration of PRP offers comparable benefit as epidural steroid injection (ESI) in the management of radiculopathy due to LDD. | ○ | Article | Preoperative controlling nutritional status score for patients with medullary thyroid cancer | Purpose: Medullary thyroid cancer (MTC) is aggressive in nature compared with differentiated thyroid cancer, with a poor 10-year overall survival rate. Given the effect of the immune-inflammatory system on the survival of patients with cancer, the aim of this study was to investigate the prognostic significance of the preoperative Controlling Nutritional Status (CONUT) score in patients with MTC. Methods: This multicenter, retrospective study included 221 patients with MTC who underwent thyroidectomy at four tertiary hospitals. The CONUT score was calculated for all patients using preoperative serum albumin and total cholesterol concentrations and lymphocyte counts. Patients were stratified into high and low CONUT score groups based on a cut-off score of 2. Results: Overall, 164 (74%) and 57 (26%) patients were categorized into low and high CONUT score groups, respectively. There were no significant differences in the clinicopathological characteristics, including age, sex, primary tumor size, preoperative levels of calcitonin and carcinoembryonic antigen, and neck lymph node and distant metastasis between the two groups. The 10-year disease-specific survival rate in the high CONUT score group was significantly shorter than that in the low CONUT score group (85.7% vs. 97.3%; p < 0.001). Multivariate analysis revealed that a high CONUT score was an independent prognostic factor for disease-specific survival (p = 0.008). Conclusion: The preoperative CONUT score appears to be an independent prognostic indicator in patients with surgically treated MTC, with a high score indicating a poorer prognosis. | ○ | ○ | Article | Gossypetin Alleviates DSS-induced Colitis by Regulating COX2 and ROS-JNK Signaling | Background Inflammatory Bowel Disease (IBD) represents a chronic and recurrent inflammatory condition affecting the gastrointestinal tract, with a rising global incidence. Current treatment approaches include surgery and drugs. However, surgeries are invasive procedures, while drug treatments often present with various side effects. Gossypetin, a flavonoid found abundantly in plants such as hibiscus, exhibits anti-oxidant and anti-cancer properties. However, its potential impact on IBD remains unexplored.Objective This study aimed to investigate the therapeutic potential of gossypetin on colitis.Methods We employed the DSS-induced colitis model to evaluate the therapeutic potential of gossypetin on colitis. The efficacy of gossypetin was assessed within this model using the Disease Activity Index (DAI) score and histological analysis. Additionally, we utilized qRT-PCR to measure the levels of inflammatory cytokines and Superoxide Dismutase (SOD). Immunohistochemistry confirmed the expression of tight junction markers, COX-2, and phosphorylated JNK protein, normally associated with disease progression. Furthermore, Western blot analysis was conducted to examine the SOD levels and anti-apoptotic effects of gossypetin.Results In DSS-induced colitis mice, gossypetin treatment ameliorated weight loss and reduced colon length caused by DSS treatment. Additionally, gossypetin-treated groups exhibited DAI scores and reduced histological damage. Moreover, gossypetin treatment increased tight junction expression, decreased inflammatory responses, reduced ROS levels, attenuated JNK signaling, and decreased apoptosis.Conclusion Gossypetin shows therapeutic potential for mitigating the symptoms and progression of colitis by targeting ROS-JNK signaling involved in inflammation and tissue damage. | ○ | Article | Photodegradation ability of electrospun Ag–Co/polysulfone nanocomposite nanofibers against acetamiprid pesticide | Non-woven neat polysulfone (PSU) and Ag–Co/PSU nanocomposite nanofibers were prepared by electrospinning process. The electrospun non-woven neat PSU and Ag–Co/PSU nanocomposite nanofibers were characterized through scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX), Fourier transform infrared spectroscopy (FTIR) and X-ray diffractometry (XRD). The SEM study presented that PSU and Ag–Co/PSU nanocomposites nanofibers were smooth, bead less, and randomly oriented. The diameter of neat PSU and Ag-Co/PSU nanocomposite nanofibers was below 800 nm. EDX spectra illustrated the elemental composition of non-woven neat PSU nanofibers and Ag–Co/PSU nanocomposite nanofibers, which showed the presence of Ag–Co NPs (nanoparticles) in the composite nanofibers. The FTIR spectrum of Ag–Co/PSU presented a peak at 710 cm−1, which might be due to the presence of Ag/Co NPs in PSU electrospun nanofibers. The photodegradation study presented that the neat PSU and Ag–Co/PSU nanocomposite nanofibers degraded about 74% and 83% acetamiprid pesticide within 5 h, respectively. It was also found that the neat PSU degraded about 20% and 67% acetamiprid pesticide while Ag–Co/PSU nanocomposite nanofibers degraded about 25% and 78% using 0.010 and 0.030 g nanofibers, respectively. Further, it was also found that the recovered PSU and Ag–Co/PSU nanocomposite electrospun nanofibers significantly degraded the acetamiprid pesticide. The photodegradation efficiency of recovered Ag–Co/PSU (47%) was higher than that of recovered neat PSU nanofibers (28%). It means that Ag–Co/PSU nanocomposite nanofibers had superior photocatalytic activity as compared to neat PSU nanofibers. In this work, to explore the potential applications of slag, it was modified with silane coupling agent 3-aminopropyltriethoxysilane (KH-550) to improve its compatibility with polymer matrix. A series of low density polyethylene (LDPE)/modified slag (MS) composites were prepared using melt blending method. The results showed that the introduction of MS significantly improved the thermal and mechanical properties of LDPE in comparison to those of its pure form. The tensile strength, tensile modulus, and Td5% of the LDPE/MS composites with 40 wt% MS content were enhanced by 58%, 113%, and 19.9 degree celsius, respectively. Furthermore, dynamic rheological analysis revealed that a higher MS filler content resulted in a higher restriction on the movement of the LDPE molecular chains.
